Abstract Robust evidence on relationships between the human microbiome and health are critical for understanding improving condition. However, there is little information about methodological approaches to combine analyze multiple data sets. To address this gap, we conducted a scoping review of studies that sequencing from sets understand study objectives, sources selection, feature -table assembly methods, analyses. Abstract Background High-throughput sequencing measurements of the vaginal microbiome have yielded intriguing potential relationships between and preterm birth (PTB; live prior to 37 weeks gestation). However, results across studies been inconsistent. Results Here, we perform an integrated analysis previously published datasets from 12 cohorts pregnant women whose microbiomes were measured by 16S rRNA gene sequencing. Of 2039 included in our analysis, 586 went on deliver prematurely. Substantial variation these existed their definition birth, characteristics study populations, methodology. Nevertheless, a small group taxa comprised vast majority all cohorts. We trained machine learning (ML) models predict PTB composition microbiome, finding low modest predictive accuracy (0.28–0.79). Predictive was typically lower when ML one dataset predicted another dataset. Earlier (< 32 weeks, < 34 weeks) more predictable than late (34–37 weeks), both within datasets. Integrated differential abundance revealed highly significant negative association L. crispatus that consistent almost studies.
