Abstract Vat photopolymerisation 3D printing is being actively explored for manufacturing personalised medicines due to its high dimensional accuracy and lack of heat application. However, several challenges have hindered clinical translation, including the inadequate speeds, resins that give soluble matrices, need non-destructive quality control measures. In this study, first time, a rapid approach producing water-soluble vat photopolymerised matrices means non-destructively verifying their drug content were investigated. Volumetric printing, novel form photopolymerisation, was used fabricate warfarin-loaded 3D-printed tablets (printlets). Eight different formulations containing varying amounts warfarin (0.5–6.0% w/w) print two sized torus-shaped printlets within 6.5 11.1 s. Nuclear magnetic resonance (NMR) spectroscopy revealed presence only trace unreacted acrylate monomers, suggesting reaction had occurred near completion. All completely solubilised released entire load 2.5 7 h. NIR (NIRS) verify dose loaded into printlets. The partial least square regression model built showed strong linearity (R 2 = 0.980), in predicting loading test sample (RMSEP 0.205%).
