The Endocrine Society's first Scientific Statement in 2009 provided a wake-up call to the scientific community about how environmental endocrine-disrupting chemicals (EDCs) affect health and disease. Five years later, substantially larger body of literature has solidified our understanding plausible mechanisms underlying EDC actions exposures animals humans—especially during development—may lay foundations for disease later life. At this point history, we have much stronger knowledge EDCs alter gene-environment interactions via physiological, cellular, molecular, epigenetic changes, thereby producing effects exposed individuals as well their descendants. Causal links between exposure manifestation are substantiated by experimental animal models consistent with correlative epidemiological data humans. There several caveats because differences work is conducted can lead difficulties drawing broad conclusions, must continue be cautious inferring causality In second Statement, reviewed on subset topics which translational evidence strongest: 1) obesity diabetes; 2) female reproduction; 3) male 4) hormone-sensitive cancers females; 5) prostate; 6) thyroid; 7) neurodevelopment neuroendocrine systems. Our inclusion criteria studies were those predominantly past 5 deemed high quality based appropriate negative positive control groups or populations, adequate sample size design, mammalian levels range that was relevant We also focused using developmental origins model. No report excluded effect exposure. bulk results across board strengthen endocrine health-related EDCs. Based more complete principles act, including nonmonotonic dose-responses, low-dose effects, vulnerability, these findings better translated human health. Armed information, researchers, physicians, other healthcare providers guide regulators policymakers they make responsible decisions.

Numerous studies have investigated the environmental occurrence, human exposure, and toxicity of bisphenol A (BPA). Following stringent regulations on production usage BPA, several analogues been produced as a replacement for BPA in various applications. The present review outlines current state knowledge occurrence (other than BPA) environment, consumer products foodstuffs, exposure biomonitoring, toxicity. Whereas was still major analogue found most monitoring studies, BPF BPS were also frequently detected. Elevated concentrations BPAF, BPF, (i.e., similar to or greater that reported abiotic environment urine from some regions. Many exhibit endocrine disrupting effects, cytotoxicity, genotoxicity, reproductive toxicity, dioxin-like neurotoxicity laboratory studies. It is likely that a half-century ago even enthusiastic and optimistic proponents of the synthetic polymer industry (Mr. McGuire included) could not have predicted massive scale on which polymers would be manufactured used today. Ultimately, future success this will rely development sustainable polymers—materials derived from renewable feedstocks are safe in both production use can recycled or disposed ways environmentally innocuous. Meeting these criteria an economical manner cannot achieved without transformative basic research hallmark journal. In Perspective we highlight five topics—the synthesis monomers degradable polymers, chemical recycling strategies, new classes reprocessable thermosets, design advanced catalysts—that believe play vital role polymers. We also offer our outlook several outstanding challenges facing community broad area

Abstract Bisphenol A (BPA) is a well-known environmental, endocrine-disrupting chemical, and bisphenol S (BPS) has been considered safer alternative for BPA-free products. The present study aims to evaluate the impact of BPA BPS on reproductive neuroendocrine system during zebrafish embryonic larval development explore potential mechanisms action associated with estrogen receptor (ER), thyroid hormone (THR), enzyme aromatase (AROM) pathways. Environmentally relevant, low levels exposure led advanced hatching time, increased numbers GnRH3 neurons in both terminal nerve hypothalamus, expression reproduction-related genes (kiss1, kiss1r, gnrh3, lhβ, fshβ, erα), marker synaptic transmission (sv2). Low similar effects: hypothalamic kiss1, erα. Antagonists ER, THRs, AROM blocked many effects gene expression, providing evidence that those three pathways mediate actions system. This demonstrates alternatives used manufacture products are not necessarily safer. Furthermore, this first describe low-level Kiss/Kiss development. It also report multiple cellular (ERα, AROM) mediating any species.
