This completely updated revised (with many figures converted colour) retains original core concepts while expanding often simplifying details. includes new chapter prefrontal cortex, which integrates frontal hippocampal views anxiety an extensively modified personality providing basis for further developments Reinforcement Sensitivity Theory. book is essential postgraduate students researchers experimental psychology neuroscience, well all clinical psychologists psychiatrists.

Physical exercise is known to reduce anxiety, but the underlying brain mechanisms remain unclear. Here, we explore a hypothalamo-cerebello-amygdalar circuit that may mediate motor-dependent alleviation of anxiety. This three-neuron loop, in which cerebellar dentate nucleus takes center stage, bridges motor system with emotional system. Subjecting animals constant rotarod engages glutamatergic neurons drive PKCδ+ amygdalar elicit an anxiolytic effect. Moreover, challenging on accelerated rather than hypothalamic provide superimposed effect via orexinergic projection activate amygdala. Our findings reveal cerebello-limbic pathway contribute motor-triggered anxiety and be optimally exploited during physical exercise.

Histamine is a conserved neuromodulator in mammalian brains and critically involved many physiological functions. Understanding the precise structure of histaminergic network cornerstone elucidating its function. Herein, using histidine decarboxylase (HDC)-CreERT2 mice genetic labeling strategies, we reconstructed whole-brain three dimensional (3D) neurons their outputs at 0.32 × 2 μm 3 pixel resolution with cutting-edge fluorescence microoptical sectioning tomography system. We quantified density all brain areas found that fiber varied significantly among regions. The was positively correlated amount histamine release induced by optogenetic stimulation or aversive stimulation. Lastly, fine morphological 60 via sparse uncovered largely heterogeneous projection pattern individual neurons. Collectively, this study reveals an unprecedented quantitative analysis projections mesoscopic level, providing foundation for future functional study.

Overly strong fear memories can cause pathological conditions. Histamine H3 receptor (H3R) has been viewed as an optimal drug target for CNS disorders, but its role in memory remains elusive. We find that a selective deficit of H3R cholinergic neurons, not glutamatergic enhances freezing level during contextual retrieval without affecting cued memory. Consistently, genetically knocking down or chemogenetically activating neurons the ventral basal forebrain (vBF) mimics this enhanced memory, whereas augmentation is rescued by re-expressing chemogenetic inhibition vBF neurons. Spatiotemporal regulation light-sensitive rhodopsin-H3R fusion protein suggests postsynaptic H3Rs presynaptic projections dorsal hippocampus, are responsible modulating Therefore, precise modulation cell-type- and subcellular-location-specific manner should be explored

Abstract Autism spectrum disorder (ASD) is a neurodevelopmental characterized by impaired social interactions, communication deficits and repetitive behaviors. A study of autistic human subjects has identified RFWD2 as susceptibility gene for autism, patients have 3 copies the gene. The role an E3 ligase in neuronal functions, its contribution to pathophysiology ASD, remain unknown. We generated knockin mice model condition high dosage . found that heterozygous ( Rfwd2 +/− ) male exhibited core symptoms autism. showed interaction communication, increased anxiety-like behavior, spatial memory deficits, whereas female subtle but were normal anxiety-like, repetitive, These autistic-like behaviors males accompanied reduction dendritic spine density abnormal synaptic function on layer II/III pyramidal neurons prelimbic area medial prefrontal cortex (mPFC), well decreased expression proteins. Impaired rescued ETV5, one major substrates RFWD2, mPFC. findings indicate important pathogenesis

Growing evidence supports the effectiveness of deep brain stimulation (DBS) in treating various psychiatric disorders. DBS has potential to selectively stimulate specific subcortical areas thus providing high-frequency electric these regions. In this review, we provide an overview across studies investigating effects NAc humans animals discuss mechanisms underlying its clinical efficacy. We address anatomical properties discuss, particular, frequently reported differential shell core DBS. Moreover, by outlining cell types, transmitter systems (i.e., predominantly GABAergic dopaminergic systems) pathways that have been be relevant for effects, aim further elucidate neurobiological determinants Finally, since treatment are most probably also related alterations connected circuits or networks, review focusing on investigation network effects. By examining components assumed relevance context DBS, will hopefully contribute increasing our knowledge about optimizing future selection optimal targets.

Abstract The ventral anterior (VA) nucleus of the thalamus is a major target basal ganglia and closely associated with pathogenesis Parkinson’s disease (PD). Notably, VA receives direct innervation from hypothalamic histaminergic system. However, its role in PD remains unknown. Here, we assessed contribution histamine to neuronal activity motor deficits. Functional magnetic resonance imaging showed reduced patients. Optogenetic activation neurons or afferents significantly alleviated deficits 6-OHDA-induced rats. Furthermore, excited via H1 H2 receptors their coupled hyperpolarization-activated cyclic nucleotide-gated channels, inward-rectifier K + Ca 2+ -activated channels. These results demonstrate that actively compensate for Parkinsonian by biasing activity. findings suggest targeting downstream ion channels may be potential therapeutic strategy dysfunction.
