RNAi has considerable potential as a cancer therapeutic approach, but effective and efficient delivery of short interfering RNA (siRNA) to tumors remains major hurdle. It challenge prepare functional siRNA complex, target enough dose the tumor, stimulate its internalization into tumor cells release cytoplasm. Here, we show how these key barriers can be overcome with complex─comprising siRNA, cationic lipids, pH-responsive peptides─that is suited uptake enhancement via focused ultrasound (FUS). The complex provides nucleic acid encapsulation, nuclease protection, endosomal escape such that gene silencing in substantially more than obtained either equivalent lipoplexes or commercial reagents. Abstract Background Hyperthermia treatment quality is usually evaluated by thermal (dose) parameters, though hyperthermic radiosensitization effects are also influenced the time interval between two modalities. This work applies biological modelling for clinical evaluation of cervical cancer patients treated with radiotherapy plus hyperthermia calculating equivalent radiation dose (EQD RT , i.e., needed same effect alone). Subsequent analyses evaluate impact logistics. Methods Biological was performed 58 23–28 fractions 1.8–2 Gy 4–5 weekly sessions. Measured temperatures (T50) and recorded intervals sessions were used to calculate EQD using an extended linear quadratic (LQ) model LQ parameters based on extensive experimental data. Next, a 30-min (optimized logistics) as well 4‑h (suboptimal evaluated. Results Median average measured T50 41.2 °C (range 39.7–42.5 °C) 79 min 34–125 min), respectively, resulting in median total enhancement (D50) 5.5 (interquartile range [IQR] 4.0–6.6 Gy). During a microwave hyperthermia oncology treatment, the target region temperature is elevated to temperatures of 40–44 °C, which improves therapeutic effect standard radiotherapy and/or chemotherapy treatments.
