Lacto-N-triose II (LNTII), lacto-N-tetraose (LNT), and lacto-N-neotetraose (LNnT) are key neutral human milk oligosaccharides (HMOs) with significant physiological functions. In cell factory-based synthesis, glycosyltransferases rate-limiting enzymes, enhancing the activity of which is, therefore, crucial for efficient synthesis HMOs. To address this issue, we developed a high-throughput screening method based on UDP-glucose regeneration-coupled colorimetric reaction. As case study, applied directed evolution β-1,3-N-acetylglucosamine transferase in Saccharomyces cerevisiae (S. cerevisiae), resulting mutant strain that doubled LNnT production. Additionally, by metabolic flux heterologous pathway, further elevated total titer to 1.29 g/L, setting new record production S. cerevisiae. Abstract This technical review, one of five developed by the European Society for Pediatric Gastroenterology Hepatology and Nutrition (ESPGHAN) special interest group (SIG) on gut microbiota modifications (GMM), supports creation a position paper use biotic‐supplemented formulas, including those containing human milk oligosaccharides (HMOs) produced through chemical synthesis or microbial biotechnology.
