Antimicrobial resistance is a critical global challenge in the 21st century, validating Sir Alexander Fleming's warning about misuse of antibiotics leading to resistant microbes. With dwindling arsenal effective antibiotics, it imperative concentrate on alternative antimicrobial strategies. Previous studies have not comprehensively discussed advantages and limitations various strategies, including bacteriophage therapy, probiotics, immunotherapies, photodynamic essential oils, nanoparticles peptides (AMPs) within single review. This review addresses that gap by providing an overview these non-antibiotic highlighting their pros cons, with particular emphasis (AMPs). We explore mechanism action AMPs against bacteria, viruses, fungi parasites. While hold significant promise, application mainstream drug development hindered challenges such as low bioavailability potential toxicity. However, advancements peptide engineering chemical modifications offer solutions enhance clinical utility. Additionally, this presents updates strategies aimed at improving cost, stability selective toxicity through peptidomimetics. These molecules demonstrated activity broad range pathogens, making them valuable candidates for integration into surface coatings prevent device-associated infections. Furthermore, we discuss approaches attaching functionalising surfaces. Finally, recommend comprehensive vivo evaluate efficacy mimetics, investigate synergistic combinations other assess medical devices.

Fungal infections are a significant global health challenge, causing approximately 3.8 million deaths annually, with immunocompromised populations particularly at risk. Traditional antifungal therapies, including azoles, echinocandins, and polyenes, face limitations due to rising resistance, toxicity, inadequate treatment options. This review explores innovative strategies for preventing managing fungal infections, such as vaccines, peptides, nanotechnology, probiotics, immunotherapy. Vaccines offer promising avenues long-term protection, despite difficulties in their development complexity immune evasion mechanisms. Antifungal peptides provide novel class of agents broad-spectrum activity reduced resistance risk, whilst nanotechnology enables targeted, effective drug delivery systems. Probiotics show potential vulvovaginal candidiasis, by maintaining microbial balance. Immunotherapy leverages system modulation enhance defenses, omics technologies deliver comprehensive insights into biology, paving the way therapeutic vaccine targets. While these approaches hold immense promise, challenges cost, accessibility, translational barriers remain. A coordinated effort among researchers, clinicians, policymakers is critical advancing addressing burden effectively.

In light of rising public health threats like antifungal and antimicrobial resistance, alongside the slowdown in new development, biomimetics have shown promise as therapeutic agents. Multidrug-resistant fungi pose significant challenges they quickly develop making traditional antifungals less effective. Developing is also complicated by need to target eukaryotic cells without harming host. This review examines biomimetic materials that mimic natural biological mechanisms for targeted efficient action. It covers a range agents, including peptides, alginate-based antifungals, chitosan derivatives, nanoparticles, plant-derived polyphenols, probiotic bacteria. These agents work through such disrupting cell membranes, generating reactive oxygen species, inhibiting essential fungal processes. Despite their potential, remain terms ensuring biocompatibility, optimizing delivery, overcoming potential resistance. Production scalability economic viability are concerns. Future research should enhance stability efficacy these materials, integrate multifunctional approaches, sophisticated delivery systems. Interdisciplinary efforts needed understand interactions between cells, host environment. Long-term environmental impacts, resistance mechanisms, standardized testing protocols require further study. conclusion, while represent revolutionary approach combating multidrug-resistant fungi, extensive development fully realize potential.
