Sebaceous free fatty acids are metabolized by multiple skin microbes into bioactive lipid mediators termed oxylipins. This study investigated correlations between oxylipins and on the superficial of pre-pubescent children (N = 36) adults 100), including pre- 25) post-menopausal females 25). Lipidomics metagenomics revealed that Malassezia restricta positively correlated with oxylipin 9,10-DiHOME adult negatively its precursor, 9,10-EpOME, skin. Co-culturing keratinocytes demonstrated a link pro-inflammatory cytokines IL-1β IL-6 production. We also observed strong other microbial taxa, highlighting life stage differences in sebum production community composition. Our findings imply complex host-microbe communication system mediated metabolism occurring human skin, warranting further research role health disease paving way towards novel therapeutic targets treatments.

To identify biomarkers and pathways to Type-2 diabetes (T2D), a major global disease, we completed array-based epigenome-wide association in whole blood 5,709 Asian people. We found 323 Sentinel CpGs (from 314 genetic loci) that predict future T2D. The reveal coherent, nuclear regulatory disturbances canonical immune activation pathways, as well metabolic networks involved insulin signalling, fatty acid metabolism lipid transport, which are causally linked development of have potential clinical utility biomarkers. An composite Methylation Risk Score (MRS) is predictive for T2D (RR: 5.2 Q4 vs Q1; P=7x10 -25 ), additive risk. Targeted methylation sequencing revealed multiple additional predicting T2D, synthesis sequencing-based MRS strongly 8.3 P=1.0x10 -11 ). Importantly, varies between ethnic groups, way explains large fraction the difference risk populations. thus provide new insights into precede sequence-based DNA markers inform stratification prevention.

Abstract Precision medicine (PM) research in recent years has witnessed a remarkable surge large-scale population genomics programs. In 2017, Singapore initiated the National Medicine (NPM) program, three-phase national strategy driving PM research, innovation, and enterprise capitalizing on Singapore’s unique multi-ancestral Asian population. Coordinated by Health Research, (PRECISE), NPM Phase I assembled an reference genome of 10,000 individuals completed using highly manual laborious laboratory sample workflows. II, targeting 100,000 cohort (PRECISE-SG100K), required transformation sequencing workflow to production-scale operation leveraging high-throughput automation robotics. Here we share detailed technical blueprints for implementing maintaining PRECISE-SG100K Genomic Engine , learning from overcoming I’s pain-points bottlenecks. The allowed successful scale up genomic throughput, enabling anticipated completion whole genomes three years.
