Methionine restriction (MetR) extends lifespan in various organisms, but its mechanistic understanding remains incomplete. Whether MetR during a specific period of adulthood increases is not known. In Drosophila, reported to extend only when amino acid levels are low. Here, by using an exome-matched holidic medium, we show that decreasing Met 10% Drosophila with or without total levels. the first four weeks adult life robustly lifespan. young flies induces expression many longevity-related genes, including sulfoxide reductase A (MsrA), which reduces oxidatively-damaged Met. MsrA induction foxo-dependent and persists for two after cessation diet. Loss attenuates extension early-adulthood MetR. Our study highlights age-dependency organismal response nutrients suggests nutrient particular sufficient healthspan extension.

Developmental transitions are often triggered by a neuroendocrine axis and can be contingent upon multiple organs achieving sufficient growth maturation. How the neurodendocrine senses size maturity of peripheral is not known. In Drosophila larvae, metamorphosis sharp increase in level steroid hormone ecdysone, secreted prothoracic gland (PG). Here, we show that BMP2/4 ortholog Dpp function as systemic signal to regulate developmental timing. from tissues, mostly imaginal discs, reach PG inhibit ecdysone biosynthesis. As discs grow, reduced signaling observed, consistent with possibility functions checkpoint mechanism prevents when insufficient. Indeed, starvation early third larval instar, reducing abrogates critical-weight which normally pupariation under these conditions. We suggest increased local trapping morphogen within tissues they grow would reduce circulating levels hence provide readout their status.
