Obezite, genetik, çevresel faktörler ve bunların karmaşık etkileşimleri tarafından yönetilen birçok kronik hastalık ile ilişkilidir. Genetiğin obeziteye yatkınlıkta önemli rol oynadığı riskine %70'e kadar katkıda bulunabileceği tahmin edilmektedir. Yapılan genetik çalışmalar, genlerin bulunduğunu doğrulamıştır. Genlerdeki anomaliler, Bardet- Biedl Prader-Willi sendromları gibi obezite ile ilişkili bozukluklardan doğrudan sorumludur. Genetik epidemiyolojik yaklaşımlar, özellikle genom çapında ilişkilendirme çalışmaları, insan obezitesinde önemli oynayan çok sayıda geni tanımlamıştır. Gelecekte, genetiğinin daha fazla araştırılması, tedavisi için yararlı teşhis testleri geliştirmeye yardımcı olabilir. Bu derleme genetiği hakkında güncel literatür desteklenmiş bilgiler aktarılmıştır.

Cortical layer 6b is considered by many to be a remnant of the subplate that forms during early stages neocortical development, but its role in adult not well understood. Its neuronal complement has only recently become subject systematic studies, and axonal projections synaptic input structures have remained largely unexplored despite decades research into function. In recent years, however, (L6b) attracted increasing attention functional beginning elucidated. this review, I will attempt provide an overview what currently known about excitatory inhibitory neurons layer, their pre- postsynaptic connectivity, implications. Similarities differences between different cortical areas highlighted. Finally, are highly responsive several neuropeptides such as orexin/hypocretin, neurotensin cholecystokinin, some cases exclusively. They also strongly controlled neurotransmitters acetylcholine norepinephrine. The interaction these neuromodulators with L6b microcircuitry consequences discussed.

Alcohol use disorders (AUDs) impose an enormous societal and financial burden, world-wide, alcohol misuse is the 7th leading cause of premature death1. Despite this, there are currently only 3 FDA approved pharmacological treatments for treatment AUDs in United States. The neurotensin (Nts) system has long been implicated modulating behaviors associated with misuse. Recently, a novel compound, SBI-553, that biases action Nts receptor 1 (NTSR1) activation, shown promise preclinical models psychostimulant Here we investigate efficacy this compound to alter ethanol-mediated comprehensive battery experiments assessing ethanol consumption, behavioral responses ethanol, sensitivity metabolism. Additionally, investigated behavior avoidance cognitive assays monitor potential side effects SBI-553. We find SBI-553 reduces binge-like consumption mice without altering or object recognition. also observe sex-dependent differences physiological sequential injections mice. In rats, show attenuates interoceptive (using Pavlovian drug discrimination task). Our data suggest targeting NTSR1 signaling may be promising attenuate misuse, adds body literature suggests common downstream target involved psychoactive multiple reinforcing substances.
