Inflammatory bowel disease (IBD) is a chronic and recurrent disease. It has been observed that the incidence prevalence of IBD are increasing, which consequently raises risk developing colon cancer. Abstract Background Inflammatory bowel disease (IBD) is a global health problem in which gut microbiota dysbiosis plays pivotal pathogenic role. Mesenchymal stem cells (MSCs) therapy has emerged as prospective novel tool for managing IBD, and can also regulate the composition of microbiota. However, functional significance MSCs-induced changes microbiome poorly understood. Methods Here, we investigated first time role mediating protective effect human umbilical cord MSCs (HUMSCs) on DSS-induced colitis. Gut alteration short-chain fatty acids (SCFAs) production were analyzed through 16S rRNA sequencing targeted metabolomics. Spectrum antibiotic cocktail (ABX), fecal transplantation (FMT) sterile filtrate (SFF) employed to evaluate intestinal flora its metabolites. Cytokine microarray, Enzyme-linked immunosorbent assay (ELISA), flow cytometry conducted assess CD4 + T homeostasis.
