Purpose: To investigate the role of S100A8/A9 in pathogenesis Sjögren's dry eye disease (SjDED) and explore its potential mechanism action. Methods: expression was determined by western blot quantitative real-time polymerase chain reaction (qRT-PCR). Tear secretion, corneal fluorescein staining, hematoxylin eosin staining were used to evaluate effect paquinimod, a inhibitor, on nonobese diabetic (NOD) mice. T helper 2 (Th2) responses protect against pathogens while also driving allergic inflammation, yet how large-scale Th2 are generated in tissue context remains unclear. Here, we used quantitative imaging to investigate early differentiation within lymph nodes (LNs) following cutaneous allergen administration. Contrary current models, observed extensive activation and "macro-clustering" of cells with migratory type-2 dendritic (cDC2s), generating specialized Th2-promoting microenvironments. Macro-clustering was integrin-mediated promoted localized cytokine exchange among reinforce differentiation, which contrasted the behavior during Th1 responses.
