Abstract Background and aims The human gut is home to a largely underexplored microbiome component, the archaeome. Little known of impact geography, urbanization, ethnicity, diet on archaeome in association with host health. We aim delineate variation healthy individuals its environmental factors homeostasis. Methods Using metagenomic sequencing, we characterized fecal archaeomes 792 adult subjects from 5 regions China, spanning 6 ethnicities (Han, Zang, Miao, Bai, Dai, Hani), consisting both urban rural residents for each ethnicity. In addition, sampled 119 variables (including lifestyle, diet, blood parameters) interrogated influences those factors, individually combined, variations. Results Population geography had strongest composition, followed by dietary habit, Overall, metadata cumulative effect size 11.0% variation. Urbanization decreased α-diversity (intrinsic microbial diversity) β-diversity (inter-individual dissimilarities) archaeome, archaea-to-bacteria ratios feces, whereas were enriched Methanobrevibacter smithii feces. Consumption buttered milk tea (a characteristic Zang population) was associated increased abundance M. . at central hub archaeal-bacterial interactions microecology, where it positively correlated abundances multitude short chain fatty acid (SCFA)-producing bacteria Roseburia faecis , Collinsella aerofaciens Prevotella copri ). In recent years, our understanding of archaeal diversity has greatly expanded, especially with the discovery new groups like Asgard archaea. These archaea show diverse phylogenetic and genomic traits, enabling them to thrive in various environments. Due their close relationship eukaryotes, a large number metagenomic studies have been performed on Research fine scale distribution, quantification saline aquatic sediments where they mostly occur, has, however, remained scarce. this study, we investigated depths shallow sediment cores from three distinct European environments: Baltic Sea near Hiddensee, coastal Lake Techirghiol Romania, an estuarine canal Piran, Slovenia. Based 16S rDNA amplicon sequencing, observe variation relative abundance occurrence at least seven different that are between environments depth distribution. Lokiarchaeia Thorarchaeia emerge as dominant across all sites, reaching maximal abundances 2.28 2.68% total microbial communities respectively, approx. 5.21% Hiddensee. Quantitative PCR assays provide insights into absolute Lokiarchaeia, supporting patterns distribution sediments. Co-occurrence network analysis indicates potential partners groups. Overall, study shows found stable component layers considerably diversified macro- microscales.

Redefining paradigms in the archaeal virus-host arms race DOI

Abstract Archaeal antiviral defense systems remain poorly characterized despite recent advances in understanding prokaryotic immunity. Here, we analyze 7,747 archaeal genomes, the largest and most diverse dataset to date, revealing a striking disparity system prevalence diversity compared Bacteria. Nearly one-third of genomes lack known beyond CRISPR-Cas restriction-modification (in contrast only 2.2% bacterial genomes), 31% contain systems, far below previous estimates. While many appear restricted Bacteria, several single-gene candidate (PDCs) are enriched Archaea. Phylogenetic analyses suggest that PDC-S27, PDC-S70, PDC-M05 likely originated Archaea, representing rare contributions immune repertoire. Consistent with earlier studies, our findings support existence deep evolutionary links between eukaryotic for argonautes viperins.
