Introduction. Systemic sclerosis (scleroderma) is an autoimmune disease that often causes inflammatory fibrosing lesions of the pulmonary interstitial tissue, referred to as lung disease. It associated with a significant reduction in survival and leading immediate cause systemic scleroderma patients’ death. Progression rate varies greatly. Since specific immunosuppressive antifibrotic treatment only justified case progressive lesions, predicting progression great practical importance. Aim: To develop approaches patients. Materials Methods. Our analysis included data patients observed Registry Myositis, Scleroderma Mixed Diseases (REMISSiZ). Inclusion criteria: Age 18+ years, proven scleroderma, eligibility for ACR/EULAR (2013), repeated high-resolution computed tomography lungs interval at least 1 year. All underwent 6-minute walking tests. Results Discussion. The study 44 (43 women) aged 62.9 ± 11.5 years average. average follow-up duration was 530 231 days. Radiological younger age, p=0.03, severity Raynaud’s phenomenon, p=0.032, shorter duration, p=0.038, distance covered walk test, p=0.05. functional impairment, determined by dynamics decrease blood oxygen saturation rest (p=0.003), breath shortness according Borg scale (p=0.006), initial test (p <0.001), presence topoisomerase I antibodies (anti-Scl-70, p=0.034), complaints (p=0.043) dysphagia (p=0.034). As result reverse stepwise selection variables, three independent predictors were selected from above indicators: Dysphagia, anti-Scl-70, test. Introduction. HIV infection, despite the measures taken, continues to spread throughout world. Unfavorable course of diseases is often determined by some concomitant pathologies. Among HIV-infected patients, tuberculosis most common disease forming comorbid condition named “TB/HIV” that reaches more than a half all cases newly diagnosed in regions Russia. In recent years, due increase HIV- infected patients’ life expectancy based on antiretroviral therapy, cardiovascular diseases, such as primarily infectious pericarditis, are gradually beginning hold significant place. Sepsis-induced cardiac dysfunction is a leading cause of mortality in intensive care units. However, the molecular mechanisms underlying septic cardiomyopathy remain elusive. Irisin cleaved product fibronectin type III domain-containing protein 5 (FNDC5) that protects heart from ischemia/reperfusion injury through upregulation mitochondrial ubiquitin ligase (MITOL). Gasdermin D (GSDMD)-dependent pyroptosis plays pivotal role by regulating homeostasis. whether irisin can regulate MITOL to inhibit GSDMD-dependent yet be investigated. Thus, this study was designed explore and its mechanisms. Our results demonstrate improves function against sepsis-induced reducing inflammation myocardial pyroptosis. Using siRNA vitro, revealed protective lipopolysaccharide (LPS)-induced cell mediated activation resulting inhibition Moreover, alleviated LPS-induced H9c2 suppressing IL-1β expression serum LDH CK-MB concentrations MITOL/GSDMD-dependent manner. Collectively, our data suggest treatment ameliorates activating inhibiting These findings highlight clinical relevance therapeutic potential for management dysfunction.

Accumulating evidence shows that oxidative stress and inflammation contribute to the development of cardiovascular disease. It has been suggested propolis possesses antioxidant anti-inflammatory activities. In this study, effects main flavonoids (chrysin, pinocembrin, galangin, pinobanksin) extract were researched. The results showed cellular ROS (Reactive oxygen species) levels, enzymes, Nrf2 (Nuclear factor erythroid 2-related 2) nuclear translocation, expression NQO1 (NAD(P)H:quinone oxidoreductase 1) HO-1 (heme oxygenase regulated by different concentrations individual extract, which good pro-oxidant effects. Diabetic cardiomyopathy (DCM) is a common complication of diabetes mellitus (DM). However, the mechanisms underlying DCM-induced cardiac injury remain unclear. Recently, role cyclic GMP-AMP synthase/stimulator interferon gene (cGAS/STING) signaling and pyroptosis in DCM has been investigated. Based on our previous results, this study was designed to examine impact irisin, mitochondrial ubiquitin ligase (MITOL/MARCH5), cGAS/STING dysfunction effect gasdermin D (GSDMD)-dependent pyroptosis. High-fat diet-induced mice H9c2 cells were used for geometry function or pyroptosis-related biomarker assessment at end experiments. Here, we show that impairs by increasing fibrosis GSDMD-dependent pyroptosis, including activation MITOL signaling. Our results confirmed protective irisin partially offset We also demonstrated plays pivotal pathological process pathogenesis. indicate treatment protects against injury, homeostasis, through upregulation.

Mitochondrial autophagy (or mitophagy) regulates the mitochondrial network and function to contribute multiple cellular processes. The protective effect of homeostatic mitophagy in cardiovascular diseases (CVDs) has attracted increasing attention. FUN14 domain containing 1 (FUNDC1), an identified receptor, plays essential role CVDs. Different expression levels FUNDC1 its phosphorylated state at different sites alleviate or exacerbate hypoxia ischemia/reperfusion injury, cardiac hypertrophy, metabolic damage through promotion inhibition mitophagy. In addition, can be enriched contact between mitochondria endoplasmic reticulum (ER), determining formation mitochondria-associated membranes (MAMs) that regulate calcium (Ca2+) homeostasis dynamics prevent heart dysfunction. Moreover, also been involved inflammatory such as septic cardiomyopathy. this review, we collect summarize evidence on roles exclusively various CVDs, describing interactions with organelles, involvement processes, associated signaling pathways. may become a promising therapeutic target for prevention management
