Abstract Total antioxidant capacity better characterizes the redox status of biological system than determination individual antioxidants separately. This study is first to assess total antioxidant/oxidant in plasma and urine patients with adrenal tumors. The group consisted 60 (31 women 29 men) masses, classified into three subgroups: non-functional incidentaloma, pheochromocytoma Cushing’s/Conn’s adenoma. number was set a priori based on our previous experiment (α = 0.05, test power 0.9) Plasma (TAC) increased incidentaloma patients, whereas decreased. oxidant (TOS) oxidative stress index (OSI) were significantly higher Ferric reducing potential (FRAP) decreased urine, while DPPH (2,2-diphenyl-1-picrylhydrazyl) antiradical activity only masses. In TAC, as well FRAP correlated positively methanephrine normethanephrine. Reduced levels clearly indicate reduced ability scavenge free radicals thus lack effective protection against Therefore, those are especially vulnerable damage, which can lead impaired cellular metabolism. Both biomarkers be used systemic tumor patients.

Adrenal masses are the most common of all human tumors. The role nitrosative stress and inflammation in cancer development has already been demonstrated. However, it is not known whether they involved pathogenesis adrenal aim study was to investigate a cross-talk between stress, hypoxia-inducible factor (HIF-1α) 75 patients with different types (non-functional incidentaloma, pheochromocytoma Cushing's/Conn's adenoma). The plasma concentrations total nitric oxide (NO), S-nitrosothiols, peroxynitrite nitrotyrosine activity serum myeloperoxidase (MPO) were measured spectrophotometrically, whereas interleukin 1 beta (IL-1β), tumor necrosis α (TNF-α) alpha using commercial ELISA kits. control group consisted 50 healthy people matched by age sex group. number subjects determined priori based on our previous experiment (power test = 0.9; 0.05).We found significantly higher (↑nitric oxide, ↑peroxynitrite, ↑S-nitrosothiols ↑nitrotyrosine) tumors, which accompanied increased inflammatory (↑myeloperoxidase, ↑interleukin ↑tumor α) hypoxia biomarkers. Peroxynitrite positively correlated aldosterone level. Nitrosative also associated HIF-1α. Interestingly, MPO differentiated from individuals high sensitivity specificity. Moreover, multivariate regression analysis, we showed that ONOO- IL-1β depended cortisol level, while ONOO-, HIF-1α aldosterone. Unfortunately, none assessed biomarkers studied, suggesting severity damage similar pheochromocytoma, Cushing's or Conn's adenoma.Adrenal tumors protein nitration/S-nitrosylation inflammation.

The toxic properties of ethanol are inextricably linked to oxidative stress. Despite many reports on the effects alcohol dependence blood redox homeostasis, there no data stress profile in alcohol-poisoned cases. There also diagnostic usefulness biomarkers determined post-mortem various biological fluids. This work investigates utility enzymatic and non-enzymatic antioxidant barrier, status, oxidative/nitrosative different fluids (such as blood, urine, vitreous humor, cerebrospinal fluid) study patients with acute intoxication. group included those who died due intoxication (n = 22). research showed a significant increase glutathione peroxidase activity, total ferric reducing power, tryptophan concentration only group's urine compared control. In other circulating fluids, both enzyme activities glycoxidation product concentrations were not significantly individuals overdose suddenly. We did observe connection between oxidation-reduction balance amount consumed before death. These unexpected observations may be caused by irreversible changes occurring at cellular level autolysis putrefaction. summary, use body assess homeostasis is limited analysis. Antioxidant activity (Total Capacity (TAC), Total Oxidant Status (TOS), Oxidative Stress Index (OSI)) antiradical (Radical-Scavenging Activity Assay (DPPH), Ferric-Reducing Power (FRAP)) were measured using colorimetric methods. FRAP level decreased incidentaloma (p<0.0001), (p<0.0001) adenoma while DPPH only masses (p<0.0001). Plasma TAC increased (p=0.0192), whereas (p=0.0343) decreased. TOS OSI (p<0.01) significantly higher In patients, (p=0.001; p=0.002), as well (p=0.007) (p=0.017) correlated positively normethanephrine. We are first who showed reduced radical scavenging capacity plasma/urine masses. Nevertheless, compared controls. Therefore, urinary antioxidant activities depend presence tumor. Lower levels TAC, clearly indicate ability scavenge free radicals thus lack effective protection against oxidative stress Both redox biomarkers can be used systemic tumor patients.

Due absence any chemotherapeutic or chemoprophylactic agents trastuzumab-induced side effects, study investigated potential role regular physical exercise antioxidant defenses, oxidative stress, nitrosative damage during treatment. The aimed analyze relationship between activity redox status women with therapy. We observed 50 therapy two groups: one that undertook moderately intensive supervised exercises, a second performed according recommendations patients, along third (control) group healthy women. The enzyme non-enzymatic activities were significantly higher exercised compared other participants. Pheochromocytoma may be overlooked or misdiagnosed in its early stages, resulting serious clinical consequences. To comprehensively and accurately diagnose pheochromocytoma, we established a smartphone-assisted colorimetric sensing platform for the determination of two biomarkers pheochromocytoma: epinephrine (Epi) glutathione (GSH). In this article, Co/Mn-doped oxide nanosheets (CoMnNSs) with oxidase-like laccase-like activities have been synthesized via one-step process. Compared to MnO2 (MnO2 NSs), CoMnNSs stronger catalytic activity due doping transition metal Co. Under optimal conditions, were utilized GSH Epi, respectively, assay could completed within 3 min, limits detection (LOD) 0.15 0.52 μM, respectively. Besides, practicability approach was confirmed human serum samples satisfactory recoveries. The experimental results indicated that designed sensor demonstrated excellent sensitivity selectivity. Thus, work not only widens idea visual diagnosis pheochromocytoma but also confirms effectiveness transition-metal strategy, which offers simple, economical, gentle means modulating nanozyme activity.
