Abstract Carob, Ceratonia siliqua L. (CS), is a legume well‐known for its edible pod pulp. Its seeds are used almost exclusively as source of the food additive E410. Although variety metabolites have been identified by HPLC and LC–MS analysis in CS, reports concerned with their isolation scarce. In this study, two flavonoid derivatives were isolated from methanolic extract CS seeds, namely, quercetin‐3‐ O ‐rhamnoside 4′‐ p ‐hydroxybenzoylisorhamnetin‐3,7‐di‐ ‐rhamnoside. Network pharmacology was unusually guide estimation biological potential compounds. Finally, ethyl acetate fraction standardized content HPLC. Ceratonia siliqua L. is a medicinal plant that has long been used in traditional Moroccan medicine to treat many diseases. This study aimed assess the impact of stages immature phase carob pulp (M1, M2, M3, M4, and M5) on phytochemical composition, antioxidant activity, antidiabetic activity The identification phenolic profile by HPLC-UV/MS-MS effect silico, vitro, vivo studies were carried out for extracts with high contents compounds from wild communes Timoulit (TM), Bin Elouidane (AW), Ouaouizerth (TG) province Azilal Béni Mellal-Khénifra region. results revealed gradual increase total sugar content over pulp’s ripening period, reaching value 2134 ± 56.23 mg GE/100 g fresh weight (FW) TG. three locations showed peak values polyphenol (TPC), flavonoid (TFC), condensed tannin (TCT) at M2 stage. AW had highest concentrations TPC (3819 226.4 GAE/100 FM), TFC (1034 57.08 QE/100 TCT (1472 28.46 CE/100 FM). DPPH assay (7892 296.1 TE/100 FM) FRAP (278.2 7.85 both demonstrated TG zone highly potent zone. In contrast, site exhibited markedly elevated 725.4 103.6 FM ABTS assay. HPLC-UV-MS/MS analysis methanolic (MEICP) areas contained several different chemical compounds. most prevalent 3-O-p-coumaroyl-5-O-caffeoylquinic acid, quercetin 3-methyl ether, gallic galloylquinic acid. Immature extract (ICPE) strongest vitro inhibition pancreatic α-amylase (IC50 = 0.405 µg/mL) against intestinal α-glucosidase 0.063 µg/mL). vivo, AW, TG, TM significantly reduced postprandial glycemia rats, having greatest effect.
