Reductase expression is a potential indicator of cellular pathology. Single-detection systems for reductases have been developed, however, the development dual-detection remain largely unexplored. We rationally designed dual-lock fluorescent probe that exhibited high signal-to-noise ratio with fluorescence Off-On response exclusively simultaneous activity two reductases, NTR and hNQO1, which are overexpressed in cancer hypoxia. The system comprised naphthalimide fluorophore control mediated by PET ICT, trimethyl-locked quinone group sensitive to nitrobenzyl carbamate NTR. This study employed hypoxia model HeLa cells demonstrate our developed detected more effectively than single-detection systems. Moreover, it enabled noninvasive real-time monitoring zebrafish embryos. Consequently, probe, strategically provides only when both NQO1 active, offers novel diagnostic platform vitro vivo applications, detecting various pathological states.
