Systemic autoinflammatory diseases (SAIDs) are a growing group of disorders caused by dysregulation the innate immune system leading to episodes systemic inflammation. In 1997, MEFV was first gene identified as disease causing for Familial Mediterranean Fever, most common hereditary SAID. cases, have strong genetic background with mutations in single genes. Since 1997 more than 30 new genes associated been identified, affecting different parts system. Macrophages can be polarized into classically activated macrophages (M1) and alternatively (M2) in the immune system, performing pro-inflammatory anti-inflammatory functions, respectively. Human THP-1 mouse RAW264.7 cell line models have been widely used various macrophage-associated studies, while similarities differences protein expression profiles between two macrophage are still largely unclear. In this study, of M1 M2 phenotypes from both were systematically investigated using mass spectrometry-based proteomics. By quantitatively analyzing more than 5,000 proteins among different types (M0, M2) lines, we identified a list that uniquely up-regulated each type further confirmed 43 commonly lines. These results revealed considerable divergences polarization macrophages. Moreover, mRNA CMPK2, RSAD2, DDX58, DHX58 strongly for models. data serve as important resources studies diseases experimental pathology human

Abstract: Rheumatoid arthritis (RA) is one of the most prevalent autoimmune diseases and a prototypic inflammatory disease, affecting small joints hands feet. Chemokines chemokine receptors play critical role in RA pathogenesis via immune cells recruitment. Several chemokines are abundant peripheral blood local inflamed RA. Furthermore, synthetic biologics disease modifying anti rheumatic drugs have been reported to affect expression. Thus, many studies focused on targeting receptors, where some shown positive promising results. However, blockers human trials treatment displayed failures that can be attributed several reasons their structures binding affinities. Nevertheless, will continue under development, order improve therapeutic potentials other diseases. In this review we provide an up-to-date knowledge regarding with emphasis activities cells. We also discussed effects those molecules This might impetus for developing new modalities treat chronic disease. Keywords: rheumatoid arthritis, chemokines, immunotherapy, immunotargets,

Acute lung injury (ALI) is a life-threatening inflammatory disease owing to the lack of specific and effective therapies. Oridonin (Ori) an active diterpenoid isolated from Rabdosiarubescens (R.rubescens) that has been shown possess broadspectrum pharmacological properties including anti-inflammatory, antitumour, antioxidative neuroregulatory effects. However, its potential protective mechanism in ALI not well characterized. In this study, we demonstrated Ori reduces mortality mice with induced by high dose lipopolysaccharide (LPS), which suggests effect on LPS ALI. Next, our results confirmed improves LPS-induced localized pulmonary pathology decreased concentration pro-inflammatory cytokines (IL-1β, IL-6, TNF-α) serum. Nuclear factor-kappa B (NF-κB) capable regulating transcription factors. Interestingly, showed inhibits expression TLR4/MyD88 phosphorylation NF-κB p65 tissues. To confirm this, further validated possible regulatory anti-inflammatory mechanisms vitro. RAW264.7 cells, are widely used as inflammation model evaluate drugs vitro, were chosen for study. Similar observed, is, pre-treatment Ori, markedly inhibited nuclear translocation subsequently release increased LPS. Overall, these exerts therapeutic inhibiting cytokines, such IL-1β, TNF-α, through TLR4/MyD88/NF-κB axis.
