Objective To identify differences between the composition, abundance, and biological function of intestinal microbiome patients with without lymph-vascular invasion (LVI) colorectal cancer (CRC) to construct predictive labels support accurate assessment LVI in CRC. Method 134 CRC were included, which divided into two groups according presence or absence LVI, their microbiomes sequenced by 16SrRNA analyzed for differences. The transcriptome sequencing data 9 transformed immune cells abundance matrix CIBERSORT algorithm, correlation among LVI-associated differential microbiomes, cells, immune-related genes GO items KEGG pathways analyzed. A random forest (RF) eXtreme Gradient Boosting (XGB) model constructed predict based on microbiome. Result There was no significant difference α-diversity β-diversity (P > 0.05). Linear discriminant analysis Effect Size (LEfSe) showed 34 enriched group 5 significantly non-lymph-vascular (NLVI) group. RF XGB prediction models top 15% ranked feature significance had good efficacy. Conclusions are 39 flora different species NLVI groups. g:Alistipes.s:Alistipes_indistinctus is closely associated vascular invasion. may be involved regulating infiltration influencing expression genes. influence process through a number potential functions. microbial markers gut can used CRC-LVI conditions.

Abstract Background Gut microbiota modulation has been demonstrated to be effective in protecting patients against detrimental effects of anti-cancer therapies, as well improve the efficacy certain treatments. Among most characterized probiotics, Lactobacillus rhamnosus GG (LGG) is currently utilized clinics alleviate diarrhea, mucositis or intestinal damage which might associated with several triggers, including Clostridium difficile infections, inflammatory gut diseases, antibiotic consumption, chemotherapy radiation therapy. Here, we investigate whether LGG cell-free supernatant (LGG-SN) exert anti-proliferative activity toward colon cancer and metastatic melanoma cells. Moreover, assess potential adjuvant effect LGG-SN combination drugs. Methods alone either 5-Fuorouracil Irinotecan was used treat human cell lines. Dimethylimidazol-diphenyl tetrazolium bromide assay employed detect cellular viability. Trypan blue staining, anti-cleaved caspase-3 anti-total versus PARP western blots, annexin V/propidium iodide flow cytometry analyses were death. Flow measurement DNA content (with propidium staining) together qPCR analysis cyclins expression cycle. Results We demonstrate that able selectively reduce viability cells a concentration-dependent way. While does not any on control fibroblasts. In cells, reduction apoptosis induction, but mitotic arrest G2/M phase Additionally, sensitizes both 5-Fluorouracil Irinotecan, thereby showing positive synergistic action. Conclusion Overall, our results suggest may contain one more bioactive molecules sensitize chemotherapeutic Thus, could proposed an ideal candidate for ground-breaking integrated approaches oncology, chemotherapy-related side overcome resistance relapse issues, thus ameliorating therapeutic response patients.

ABSTRACT Colorectal cancer (CRC) screening and early diagnosis is an effective strategy for reducing CRC mortality. However, the current detection methods involve exorbitant costs complex procedures, which are inconvenient large‐scale screening. Given its high prevalence in malignant tissues feces of patients, Fusobacterium nucleatum ( F. ) has emerged as a crucial biomarker CRC. Herein, we propose ‐specific recognition diagnosis. A novel nanobioprobe (AIE‐Pep) with aggregation‐induced emission (AIE) characteristics was synthesized by conjugating red/near‐infrared (NIR) emissive AIE luminogen (AIEgen) FadA‐targeting peptide (ASANWTIQYND). The robust binding affinity between FadA on allows AIE‐Pep NPs to adhere selectively , emits strong red/NIR fluorescence. In model orthotopic CRC, can precisely localize around Moreover, demonstrated limit (LOD) 82.97 CFU/mL could significantly differentiate mice from those normal mice. Overall, this study presents pivotal approach specifically identifying holds immense potential

Gut microbiota plays a crucial role in modulating immune responses, including effector response to infection and surveillance of tumors. This article summarizes the current scientific evidence on effects supplementation with prebiotics, probiotics, synbiotics high-risk human papillomavirus (HPV) infections, precancerous lesions, various stages cervical cancer development treatment while also examining underlying molecular pathways involved. Our findings indicate that higher dietary fiber intake is associated reduced risk HPV infection, certain probiotics have shown promising results clearing HPV-related lesions. Additionally, strains prebiotics such as inulin fructo-oligosaccharides, decrease frequency gastrointestinal adverse patients. These agents attain their by metabolic pathways, reduction inflammation oxidative stress, promoting apoptosis, inhibiting cell proliferation, suppressing activity oncogenes, thus attenuating tumorigenesis. We conclude although further studies are necessary, robust preclinical models demonstrates play an essential cancer, from carcinogenesis its medical treatment. Consequently, we strongly recommend conducting high-quality clinical trials using these adjuvants since they proven safe.
