Purpose: Neurodegenerative diseases are associated with neuroinflammation along activation of microglia and oxidative stress, but currently lack effective treatments. Punicalagin is a natural bio-sourced product that exhibits anti-inflammatory effects on several chronic diseases; however, the anti-oxidative have not been well examined. This study aimed to investigate punicalagin LPS-induced inflammatory responses, NLRP3 inflammasome activation, production ROS using murine BV2 cells. Multiple sclerosis (MS) is a progressive neuro-inflammatory and neuro-autoimmune disease. Although hydrogen sulfide has recently shown potential therapeutic impacts in different neurological diseases, its effects on MS are still obscure. MiR-146a considered vital target for approaches treating MS. The present study directed to explore the of NaHS (hydrogen donor) cuprizone-induced whether can mediate via regulating miR-146a expression. A total 28 male C57Bl/6 mice were divided into 4 groups; control, cuprizone-intoxicated, control (100 μmol/kg/day, i.p), NaHS-treated groups. Intriguingly, treatment managed improve locomotor coordination curb neuronal inflammation demyelination as evidenced by hematoxylin & eosin, Luxol fast blue staining increased myelin basic protein (MBP) content. Additionally, reduced interleukin-1 receptor-associated kinase-1 (IRAK-1), nuclear transcription factor kappa B (NF-κB), interleukin (IL)-17, IL-1β brain levels along with downregulation expression compared untreated cuprizone-intoxicated group. Furthermore, animals revealed much less oxidative stress elevated glutathione malondialdehyde contents.
