Abstract Forkhead box O (FOXO) transcription factors regulate diverse biological processes, affecting development, metabolism, stem cell maintenance and longevity. They have also been increasingly recognised as tumour suppressors through their ability to genes essential for proliferation, death, senescence, angiogenesis, migration metastasis. Mechanistically, FOXO proteins serve key connection points allow proliferative, nutrient stress signals converge integrate with distinct gene networks control fate, metabolism cancer development. In consequence, deregulation of expression function can promote genetic disorders, metabolic diseases, deregulated ageing cancer. Metastasis is the process by which cells spread from primary often via bloodstream or lymphatic system major cause death. The regulation occur predominantly at post-transcriptional post-translational levels mediated regulatory non-coding RNAs, interactions other protein partners co-factors a combination modifications (PTMs), including phosphorylation, acetylation, methylation ubiquitination. This review discusses role in initiation progression, particular emphasis on An understanding how signalling modulate developmental, tumour-suppressive functions normal tissues will offer new perspective tumorigenesis metastasis, open up therapeutic opportunities malignant diseases.

Cell cycle regulation is orchestrated by a complex network of interactions between proteins, enzymes, cytokines, and cell signaling pathways, vital for proliferation, growth, repair. The occurrence, development, metastasis tumors are closely related to the cycle. can be synergistic with chemotherapy in two aspects: inhibition or promotion. sensitivity tumor cells chemotherapeutic drugs improved cooperation strategies. This review presented mechanism commonly used effect on tumorigenesis interaction cancer treatment was briefly introduced. current collaborative strategies discussed detail.
