ABSTRACT Vaginitis is the most common problem afflicting women of childbearing age. However, underlying etiological factors remain poorly understood, leading to recurrent vaginitis and constraining clinical management. Here, we explored whether gut microbiota influences risk by performing a two-sample Mendelian randomization analysis using largest genome-wide association studies date. Four taxa in genus levels were identified related vaginitis: Candidatus Soleaferrea (inverse-variance weighted [IVW] odds ratio [OR] = 2.20, P 0.026), Dialister (IVW OR 2.62, 0.029), Lachnospiraceae UCG-008 0.41, 0.0067), Ruminiclostridium 5 0.080, 1.42 × 10 −5 ). We further mediation effect plasma metabolites two-step (MR) multivariable MR. The findings indicated that 3-phosphoglycerate lysophosphatidylcholine antagonistically act against two ( , respectively) vaginitis, thus appearing confer protective effects vaginitis. On contrary, elevation arachidonate/pyruvate reduction palmitate/myristate mediated These support potential causal role for development thereby providing strategies its prevention intervention. IMPORTANCE Besides, human vagina are important organs both colonized thousands microorganisms impacting physiology health. Whether there an interplay between microecosystems intriguing unclear. This study evaluated relationship suggested specific types may be or suppressed certain metabolites. provide treatment insights

Background Recent studies have shown that an imbalance in gut microbiota (GM) may not always be associated with endometriosis (EMS). To investigate this further, we conducted a two-sample Mendelian randomization study. Methods MR analysis was performed on genome-wide association study (GWAS) summary statistics of GM and EMS. Specifically, the MiBioGen GWAS ( N = 18,340) used as exposure. The FinnGen (8,288 EMS cases 68,969 controls) outcome. We primarily inverse variance weighted (IVW) method to analyze correlation sensitivity verify its reliability. Tryptophan (TRP) and its indole metabolites exhibit numerous biological effects, especially their antioxidant properties. This study used untargeted metabolomics in conjunction with targeted to investigate the differential expression of tryptophan follicular fluid (FF) diminished ovarian reserve (DOR) normal (NOR) populations. included patients DOR (n = 50) females NOR 35) who received vitro fertilization embryo transfer. Untargeted suggests that affects metabolic profile FF, TRP were significantly down-regulated group. Targeted quantification revealed levels TRP, IPA IAA FF group lower than those (P < 0.01). The concentration is positively correlated available rate females. These results provide data support explore pathogenesis look for new biomarkers protectors. Additionally, alterations may indirectly reflect interaction between intestinal flora microenvironment.
