Background/Objectives: Centella asiatica (L.) Urban (family Apiaceae) (C. asiatica) is a traditional botanical medicine used in aging and dementia. Water extracts of C. (CAW) have been to treat neuropsychiatric symptoms related animal models are associated with increases antioxidant response element (ARE) genes improvements mitochondrial respiratory function neuronal health. Because multiple sclerosis (MS) shares its neurogenerative pathology oxidative stress dysfunction dementia, MS may also benefit from asiatica. To determine whether CAW similarly benefits symptoms, ARE gene expression, respiration inflammatory MS, the effects on clinical disability inflammation, we tested using experimental autoimmune encephalomyelitis (EAE). Methods: C57BL/6J mice induced EAE were treated or placebo for 2 weeks. The outcomes disability, signs anxiety (open field test), respiration, inflammation demyelination. Results: At dosing schedule concentrations tested, CAW-treated demonstrated increased expression activity compared those placebo-treated EAE. was reduced infiltrates spinal cord, but differences between populations activated versus quiescent microglia equivocal. did not improve behavioral performance, motor Conclusions: In model demonstrates similar neuroprotective it exhibits dementia mouse models. These benefits, along anti-inflammatory CAW, support further investigation people MS.

Centella asiatica (CA), known for its health-promoting properties, is rich in bioactive compounds. This study optimised ultrasound-assisted extraction (UAE) parameters to maximise total phenolic content (TPC) and flavonoid (TFC) using the response surface methodology (RSM). Ethanol concentration solvent volume significantly influenced TPC TFC yields (p < 0.0001), while ultrasonic power had nonsignificant effects 0.05). Time showed no significant effect on > 0.05) but due flavonoids’ sensitivity degradation Variable interactions were negligible The relationship between responses (TPC TFC) independent could be expressed as quadratic models fitted with a Predicted R2 of 0.8263 0.9006 TFC. Based RSM, optimal conditions—75% ethanol concentration, 87.5 W power, 30 min time, 20 mL volume—yielded values 52.29 ± 1.65 mg/g 43.71 1.92 mg/g, closely aligning model predictions at 95% confidence. Additionally, UAE condition provided asiaticoside 37.56 4.25 madecassoside 16.91 1.28 mg/g. offers valuable insights into factors influencing efficiency, sustainability, scalability recovering compounds, underscoring potential sustainable method developing functional food ingredients from CA.

Stroke is known for its high disability and mortality rates. Ischemic stroke (IS), the most prevalent form, imposes a considerable burden on affected individuals. Nevertheless, existing treatment modalities are hindered by limitations, including narrow therapeutic windows, substantial adverse effects, suboptimal neurological recovery. Clarifying pathological mechanism of IS prerequisite developing new strategies. In this context, functional disruption mitochondria, endoplasmic reticulum (ER), crosstalk mechanisms between them have garnered increasing attention their contributory roles in progression IS. Therefore, review provides comprehensive summary current pathomechanisms associated with involvement ER mitochondria IS, emphasising Ca2+ destabilization homeostasis, stress, oxidative disordered mitochondrial quality control, transfer. Additionally, article highlights interaction as well mitochondrial-ER contacts (MERCs) that structurally connect ER, aiming to provide ideas references research

Alterations in epigenetic modifications, like DNA methylation, peripheral blood could serve as a useful, minimally invasive biomarker of the effects anti-aging interventions. This study explores this potential with water extract botanical Centella asiatica (CAW). Eighteen-month-old mice were treated CAW their drinking for 5 weeks alongside vehicle-treated eighteen-month-old C57BL6 mice. Reduced representation bisulfite sequencing (RRBS) was used to identify genome-wide differential methylation CAW-treated aged compared Our results showed distinct enrichment differentially methylated regions (DMRs) nearby genes involved biological processes relevant aging (i.e., antioxidant response, metabolic regulation, cellular metabolism). A difference observed between males and females both number sites state methylation. Moreover, or overlapping DMRs found be enriched related previously described mouse brain calcium circadian rhythm). Together, our data suggest that signature readily accessible, CAW's aging.
