Abstract Small cell lung cancer (SCLC) is a highly malignant with poor prognosis. Therefore, more accurate models to identify high-risk patients are essential for facilitating personalized management of SCLC. Immunogenic death (ICD) process that stimulates robust anti-tumor immune responses and holds promising implications treatment. Abstract Glutathione synthetase (GSS) catalyzes the final step in synthesis of glutathione (GSH), a well-established antioxidant. In this study, we identified pachytene spermatocytes as primary site GSS expression and generated mouse model with postnatal deletion using Stra8-Cre (S8) investigate role germ cells. This study aims to investigate the role of ferroptosis, an iron-dependent form regulated cell death, in male infertility. The motivation behind this research stems from increasing recognition oxidative stress and iron metabolism dysregulation as critical factors reproductive health. In study, 28 infertile patients (grouped by presence urogenital infections or varicocele) 19 fertile men were selected. Spermiograms performed light microscopy (WHO, 2021). Testosterone, ferritin, transferrin-bound iron, transferrin, F2-isoprostanes (F2-IsoPs) detected seminal plasma. Glutathione peroxidase 4 (GPX4) acyl coenzyme A synthetase long chain family member (ACSL4) also assessed sperm cells using enzyme-linked immunosorbent assays (ELISA). All variables correlated (statistically significant Spearman’s rank correlations) whole population, then comparison between different groups carried out. Seminal ferritin transferrin positively with F2-IsoPs, which had positive correlations ACSL4 cells. Ferritin negatively parameters. No correlation was for GPX4. Comparing three examined groups, elevated levels observed varicocele; GPX4 similar groups. These results suggested a mechanism identified increased occurrence lipid peroxidation. Such events appear be GPX4-independent pathologies such varicocele infections.
