Uveal melanoma (UM) is the most common intraocular malignant tumor and frequent not affecting skin. While rate of UM occurrence relatively low, about 50% patients develop metastasis, primarily to liver, with lethal outcome despite medical treatment. Notwithstanding that etiopathogenesis still under investigation, a set known mutations chromosomal aberrations are associated its pathogenesis have relevant prognostic value. The frequently mutated genes BAP1, EIF1AX, GNA11, GNAQ, SF3B1, mutually exclusive occurring in GNAQ almost ones BAP1 EIF1AX. Among aberrations, monosomy chromosome 3 frequent, followed by gain 8q, full or partial loss chromosomes 1 6. In addition, epigenetic mechanisms regulated non-coding RNAs (ncRNA), namely microRNAs long RNAs, also been investigated. Several papers investigating role ncRNAs reported their dysregulated expression affects cancer-related processes both vitro vivo models. This review will summarize current findings genetic mutations, ncRNA dysregulation establishing biology.

PHI-1, encoded by PPP1R14B, regulates cellular protein phosphatase-1 (PP1) signaling and has emerged as both a biomarker therapeutic target. Initially identified phospholipase-neighboring gene (PNG), PHI-1 is now known for its phosphorylation-dependent inhibition of PP1 holoenzymes, with bi-directional roles depending on expression levels. Under physiological conditions, selectively activity to maintain homeostasis, whereas pathological upregulation promotes oncogenic pathways, stabilizes tumor-promoting proteins, modulates immune responses. This article explores PHI-1’s emerging role pan-cancer in parallel emphasizing functions networks, smooth muscle contraction, cytoskeletal dynamics, selective proteostasis. The mechanistic insights highlight potential precision oncology, offering opportunities developing diagnostics therapies that target conditional functions.
