Knowledge on the myeloproliferative neoplasms (MPNs) – polycythemia vera (PV), essential thrombocythemia (ET), primary myelofibrosis (PMF) has accumulated since discovery of JAK/STAT-activating mutations associated with MPNs: JAK2 V617F, observed in PV, ET and PMF; MPL CALR mutations, found PMF. The intriguing lack disease specificity these chronic inflammation MPNs, triggered a quest for finding what precisely determines that MPN patients develop or PMF phenoptype. mechanisms action MPN-driving concomitant ( ASXL1, DNMT3A, TET2 , others), have been extensively studied, as well role played by inflammation, several pathogenic models proposed. In parallel, different types drugs tested MPNs (JAK inhibitors, interferons, hydroxyurea, anagrelide, azacytidine, combinations those), some acting both inflammation. Yet remain incurable diseases. This review aims to present current, detailed knowledge specifically may pave way development novel, curative therapies.

Osteoarthritis (OA) is a chronic musculoskeletal disorder characterized by cartilage degeneration and synovial inflammation. Paracrine interactions between chondrocytes macrophages play an essential role in the onset progression of OA. In this study, replicating inflammatory response during OA pathogenesis, were treated with interleukin-1β (IL-1β), lipopolysaccharide interferon-γ. addition, coculture system was developed to simulate biological situation joint. we examined impact hyaluronic acid (HA) viscosupplement, particularly Hyruan Plus, on macrophages. Notably, viscosupplement has demonstrated promising outcomes reducing inflammation; however, underlying mechanism action remains elusive. The attenuated inflammation, showing inhibitory effect nitric oxide production, downregulating proinflammatory cytokines such as matrix metalloproteinases (MMP13 MMP3), upregulating expression levels type II collagen aggrecan chondrocytes. HA also reduced level IL-1β, TNF-α, IL-6 macrophages, exerted overall protective partially suppressing MAPK pathway p65/NF-κB signaling Therefore, shows potential for treating arthritic joints.

In immunology, the role of macrophages extends far beyond their traditional classification as mere phagocytes; they emerge pivotal architects immune response, with function being significantly influenced by multidimensional environmental stimuli. This review investigates nuanced mechanisms which diverse external signals ranging from chemical cues to physical stress orchestrate macrophage polarization, a process that is crucial for modulation responses. By transitioning between pro-inflammatory (M1) and anti-inflammatory (M2) states, exhibit remarkable plasticity, enabling them adapt influence surroundings effectively. The exploration polarization provides compelling narrative on how these cells can be manipulated foster an environment conducive tissue repair regeneration. Highlighting cutting-edge research, this presents innovative strategies leverage dynamic interplay environment, proposing novel therapeutic avenues harness potential in regenerative medicine.
