ABSTRACT Peganum harmala, a member of the Zygophyllaceae family, is known for its diverse biological activities, including anti‐inflammatory properties. The mechanisms through which P. harmala polysaccharide (LTP) induces autophagy, however, remain largely unexplored. This study aims to elucidate role LTP in autophagy induction and efficacy mitigating inflammation within macrophages. Autophagosome formation was evaluated using GFP‐LC3 vectors, LC3‐II levels induced by were analyzed laser scanning confocal microscopy. Western blotting assessed expression autophagy‐related proteins phosphorylation state p70S6K NRK cells, treated both with without LTP, alongside inducers inhibitors. Additionally, RAW264.7 cells 1 μg/mL lipopolysaccharides (LPS), followed ELISA assays quantify inflammatory markers. study's outcomes demonstrate that facilitates an increase autophagic activity, as evidenced enhanced reduced p62 effect mediated activation AMPK‐mTOR signaling pathway, inhibiting autophagosome–lysosome fusion process. In vitro experiments LPS showed markedly decreased TNF‐α IL‐6. Our findings indicate effectively reduces LPS‐stimulated macrophages promoting via mTOR‐dependent mechanism.

In plants, 5mC DNA methylation is an important and conserved epistatic mark involving genomic stability, gene transcriptional regulation, developmental abiotic stress response, metabolite synthesis, etc. However, the roles of modification (5mC methylation) in tea plant growth development (in pre-harvest processing) flavor substance synthesis pre- post-harvest processing are unknown. We therefore conducted a comprehensive analysis four key tissues (root, leaf, flower, fruit) two processed leaves during oolong processing. found that differential among closely related to tissue functional differentiation genes expressed tissue-specifically, responsible for tissue-specific functions, maintain relatively low levels relative non-tissue-specifically genes. Importantly, hypomethylation modifications CsAlaDC TS/GS roots provided molecular basis dominant theanine roots. addition, integration methylationomics, metabolomics, transcriptomics revealed content changes metabolites were associated with transcription level corresponding genes, transcript these strictly regulated by methylation. further report some methylation, which can effectively explain aroma metabolites, including α-farnesene, nerolidol, lipids, taste substances such as catechins. Our results not only highlight processing, but also provide epimutation-related targets future improvement quality or breeding whole-tissue high-theanine varieties.

Herpes simplex virus (HSV) infections are highly widespread among humans, producing symptoms ranging from ulcerative lesions to severe diseases such as blindness and life-threatening encephalitis. At present, there no vaccines available, some existing antiviral treatments can be ineffective or lead adverse effects. As a result, is need for new anti-HSV drugs. In this report, the in vitro effect of 9,9′-norharmane dimer (nHo-dimer), which belongs β-carboline (βC) alkaloid family, was evaluated. The exhibited virucidal properties did not impede either attachment penetration steps viral particles. only exerted under constant presence incubation media, mechanism action found involve later events infection. Analysis fluorescence lifetime imaging data showed that nHo-dimer internalized well into cells when present extracellular medium, with preferential accumulation perinuclear organelles including mitochondria. After washing host fresh medium free nHo-dimer, signal decreased, suggesting partial release compound cells. This agrees observation solely manifested consistently media.
