Activated immune-inflammatory pathways insulin resistance significantly contribute associated In a cohort 90 individuals, categorized into those without LC, we evaluated, 3-6 months following acute SARS-CoV-2 infection, correlations between Hamilton Depression (HAMD), Anxiety (HAMA), Fibro-Fatigue (FF) Rating Scale scores, serum C-reactive protein (CRP), prostaglandin E2 (PGE2), galanin-galanin receptor 1 (GAL-GALR1) signaling, resistance, insulin-like growth factor (IGF-1), plasminogen activator inhibitor-1 (PAI1), S100B neuron-specific enolase (NSE). Chronic pain is a debilitating symptom with significant negative impact on the quality of life and socioeconomic status, particularly among adults elderly. Major Depressive Disorder (MDD) stands out as one most important comorbid disorders accompanying chronic pain. The kynurenine pathway serves primary route for tryptophan degradation holds critical significance in various biological processes, including regulation neurotransmitters, immune responses, cancer development, metabolism, inflammation. This review encompasses key research studies related to context headache, neuropathic pain, gastrointestinal disorders, fibromyalgia, fatigue syndrome, MDD. Various metabolites produced pathway, such kynurenic acid quinolinic acid, exhibit neuroprotective neurotoxic effects, respectively. Recent have highlighted involvement its pathophysiology Moreover, pharmacological interventions targeting shown therapeutic promise management. Abstract Background Persistent infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), reactivation of dormant viruses, and immune-oxidative responses are involved in Long COVID. Objectives To investigate whether COVID depressive, anxiety chronic fatigue (CFS) symptoms, associated IgA/IgM/IgG to SARS-CoV-2, human Herpesvirus type 6 (HHV-6), Epstein-Barr Virus (EBV), biomarkers. Methods We examined 90 patients healthy controls. measured serum against HHV-6 EBV their deoxyuridine 5′-triphosphate nucleotidohydrolase (duTPase), activin-A, C-reactive protein (CRP), advanced oxidation products (AOPP), insulin resistance (HOMA2-IR). Results showed significant elevations IgG/IgM-SARS-CoV-2, IgG/IgM-HHV-6 HHV-6-duTPase, IgA/IgM-activin-A, CRP, AOPP, HOMA2-IR. Neural network analysis yielded a highly predictive accuracy 80.6% for the diagnosis (sensitivity: 78.9%, specificity: 81.8%, area under ROC curve=0.876); topmost predictors were: IGA-activin-A, IgG-HHV-6, IgM-HHV-6-duTPase, IgG-SARS-CoV-2, IgM-HHV-6 (all positively) factor extracted from all IgA levels viral antigens (inversely). The top-5 affective symptoms due education, IgA-activin-A (predictive r=0.636). CFS were descending order: IgG-HHV-6-duTPase, IgM-activin-A, IgM-SARS-CoV-2, accuracy: r=0.709). Conclusion Reactivation HHV-6, SARS-CoV-2 persistence, autoimmune reactions activin-A combined activated pathways play major role pathophysiology as well severity

The coexistence of anxiety or depression with coronary heart disease (CHD) is a significant clinical challenge in cardiovascular medicine. Recent studies have indicated that hypothalamic-pituitary-adrenal (HPA) axis activity could be promising focus understanding and addressing the development treatments for comorbid CHD depression. HPA helps to regulate levels inflammatory factors, thereby reducing oxidative stress damage, promoting platelet activation, stabilizing gut microbiota, which enhance survival regeneration neurons, endothelial cells, other cell types, leading neuroprotective cardioprotective benefits. This review addresses relevance nervous systems, as well latest research advancements regarding its mechanisms action. discussion includes detailed function regulating processes mentioned.
