Abstract Background : Disulfidptosis is a novel form of cell death that induces disulfide stress leading to death. Therefore, this may be new direction for future cancer therapy. More and more studies have shown long non-coding RNAs (lncRNAs) can regulate gastric cancer-related biological processes. At present, there no research on disulfidptosis-related lncRNAs (DRLs) in GC. Methods The Cancer Genome Atlas (TCGA) was used retrieve information about RNA sequencing data, clinical genetic mutation data GC patients. genomic patients were downloaded from the Atlas. First, TCGA are randomly assigned training set validation set. Then, predictive risk model built using univariate multivariate Cox regression models, as well least absolute shrinkage selection operator (LASSO). value verified by receiver operating characteristic (ROC) curves concordance index (C-index) entire analysis, nomogram, correlation analysis clinicopathological characteristics confirm utility prognostic model. Abstract The context of long noncoding RNAs (lncRNAs) contains many unannotated open reading frames (ORFs). These ORFs potentially encode novel proteins or peptides with crucial roles in various human cancers, yet the translational potential these lncRNAs and functions protein products remain largely unexplored, especially gastric cancer (GC). In this study, a comprehensive analysis is performed identified GC associated lncRNA known as HCP5, which non‐canonical ORF. Further showed that HCP5‐132aa, microprotein encoded by HCP5 harboring ORF, highly expressed cells tissues, can promote proliferation inhibiting ferroptosis. Mechanistically, HCP5‐132aa enhances interaction between YBX1 ELAVL1, facilitates recognition at m 5 C site 3′UTR SLC7A11 G6PD mRNA, preserves their stability via ELAVL1. By employing Cas9/sgRNA delivery system AAV vivo, effectively knocked out inhibition tumor growth patient‐derived xenograft model are achieved. findings demonstrate derived from mediates repression ferroptosis, thereby driving progression identifying new therapeutic target for its treatment.

Background Raised erosive gastritis (REG) is a chronic with high risk of malignant transformation. Current treatments often result in recurrence rates and complications. Jianpi Qinghua Sanyu Yin (JPQHSYY), traditional Chinese medicine, shows promise treating REG. However, the underlying molecular mechanisms remain unclear. This study aimed to investigate potential mechanism JPQHSYY’s therapeutic effects on Methods RNA-seq was employed systematically analyze mRNA, lncRNA, miRNA profiles gastric mucosal tissues from REG patients before after JPQHSYY treatment. The pivotal lncRNA-miRNA miRNA-mRNA networks were predicted sequencing data bioinformatic analysis, results exported using Cytoscape software. Gene Ontology (GO) analysis Kyoto Encyclopedia Genes Genomes (KEGG) analyses used for functional exploration. Real-time quantitative reverse transcription-polymerase chain reaction (qRT-PCR) performed validate results. CCK8, cell cycle, apoptosis western blot detect miR-122-5p GES-1 cells vitro . Results revealed 576 differentially expressed lncRNAs (269 upregulated, 307 downregulated), 33 miRNAs (13 20 1717 mRNAs (777 940 downregulated) JPQHSYY-treated patients. GO KEGG highlighted key pathways, including PI3K/AKT signaling pathway, involved cycle regulation. ceRNA network suggested that impacts miRNA–lncRNA interactions. Validation experiments confirmed inhibits reducing viability, colony formation, promoting transfected cells.
