Background: Curcumin, quercetin, and vitamin D3 (cholecalciferol) are common natural ingredients of human nutrition reportedly exhibit promising anti-inflammatory, immunomodulatory, broad-spectrum antiviral, antioxidant activities. Objective: The present study aimed to investigate the possible therapeutic benefits a single oral formulation containing supplements curcumin, cholecalciferol (combinedly referred here as CQC) an adjuvant therapy for early-stage symptomatic coronavirus disease 2019 (COVID-19) in pilot open-label, randomized controlled trial conducted at Mayo Hospital, King Edward Medical University, Lahore, Pakistan. Methods: Reverse transcriptase polymerase chain reaction (RT-PCR) confirmed, mild moderate COVID-19 outpatients were receive either standard care (SOC) (n = 25) (control arm) or daily co-supplementation 168 mg 260 9 µg (360 IU) cholecalciferol, two soft capsules b.i.d. add-on SOC (CQC 14 days. includes paracetamol with without antibiotic (azithromycin). Curcuminoids (Me08 e Me23) curcumin (CUR) are derived from Curcuma Longa extract (EXT). Many therapeutic actions have been linked to these compounds, including antiviral action. Given the severe implications of especially within central nervous system, our study aims shed light on potential curcuminoids against SARS-CoV-2 infection, particularly in cells. Here, we investigated effects CUR, EXT, Me08 Me23 human neuroblastoma SH-SY5Y. We observed that significantly decreased expression plasma membrane-associated transmembrane protease serine 2 (TMPRSS2) TMPRSS11D, consequently mitigating elevated ROS levels induced SARS-CoV-2.
