Abstract Nuclear factor-kappaB (NF-ĸB) plays a crucial role in both innate and adaptive immune systems, significantly influencing various physiological processes such as cell proliferation, migration, differentiation, survival, stemness. The function of NF-ĸB cancer progression response to chemotherapy has gained increasing attention. This review highlights the inflammation control, biological mechanisms, therapeutic implications treatment. is instrumental altering release inflammatory factors TNF-α, IL-6, IL-1β, which are key regulation carcinogenesis. Specifically, conditions including colitis, upregulation can intensify inflammation, potentially leading development colorectal cancer. Its pivotal extends regulating tumor microenvironment, impacting components macrophages, fibroblasts, T cells, natural killer cells. influences tumorigenesis dampen anti-tumor responses. Additionally, modulates death notably by inhibiting apoptosis ferroptosis. It also dual stimulating or suppressing autophagy cancers. Beyond these functions, controlling stem fostering angiogenesis, metastatic potential through EMT induction, reducing sensitivity radiotherapy. Given its oncogenic capabilities, research focused on products small molecule compounds that suppress NF-ĸB, offering promising avenues for therapy.

Abstract The gut microbiome is a complex system that directly interacts with and influences many systems in the body. This delicate balance of microbiota plays an important role health disease highly influenced by lifestyle factors surrounding environment. As further research emerges, understanding full potential impact using nutraceuticals to positively influence its function may open door greater therapeutic outcomes treatment prevention disease. Curcumin, bioactive compound derived from turmeric rhizome, has been studied depth for on human as potent anti-inflammatory antioxidant properties. However, activity curcumin limited low oral bioavailability. While most available primarily focused curcuminoid compounds turmeric, non-curcuminoid hold promise offer benefits while synergistically enhancing bioavailability supporting microbiome. review summarizes current knowledge relationship between various within body, how dysbiosis, or disruption microbial balance, leads inflammation increased risk chronic also recent focuses bioactivity both comprise whole root their synergistic support healthy promising use

Abstract Colorectal cancer (CRC) development and progression, one of the most common cancers globally, is supported by specific mechanisms to escape cell death despite chemotherapy, including cellular autophagy. Autophagy an evolutionarily highly conserved degradation pathway involved in a variety processes, such as maintenance homeostasis clearance foreign bodies, its imbalance associated with many diseases. However, role autophagy CRC progression remains controversial, it has dual function, affecting either or survival, senescence tumor therapy. Indeed, numerous data have been presented that serves alternative apoptosis when latter ineffective apoptosis‐resistant cells, which why also referred programmed type II. Curcumin, active constituents Curcuma longa , great potential combat influencing various signaling pathways epigenetic regulation safe cost‐effective approach. This review discusses efficacy curcumin against vitro vivo, particularly modulation pathways. While clinical studies assessed prevention treatment, none specifically examined Nonetheless, we offer overview correlations support use this polyphenol prophylactic co‐therapeutic agent CRC.

Abstract Colorectal cancer (CRC) is a significant health problem with elevated mortality rates, prompting intense exploration of its complex molecular mechanisms and innovative therapeutic avenues. Resveratrol (RSV), recognised for anticancer effects through SIRT1 activation, promising candidate CRC treatment. This study focuses on elucidating RSV's role in progression, particularly effect autophagy‐related apoptosis. Using bioinformatics, protein imprinting immunohistochemistry, we established direct correlation between FOXQ1 adverse prognosis. Comprehensive vitro experiments confirmed ability to promote apoptosis cells. Plasmids modulation were used investigate underlying mechanisms. Molecular docking, glutathione‐S‐transferase pull‐down immunoprecipitation highlighted activation SIRT1, resulting the inhibition expression. Downstream interventions identified ATG16L as crucial autophagic target. In vivo studies validated potential therapy SIRT1/FOXQ1/ATG16L pathway. establishes capacity enhance cell CRC, positioning RSV prospective agent within

Hypoxia-inducible factor (HIF)-1 is a transcription that regulates the expression of target genes associated with oxygen homeostasis under hypoxic conditions, thereby contributing to tumor development and progression. Accumulating evidence has demonstrated HIF-1α mediates different biological processes, including angiogenesis, metastasis, metabolism, immune evasion. Thus, overexpression strongly poor prognosis in cancer patients. Natural compounds are important sources anticancer drugs studies have emphasized decisive role these mediators modulating HIF-1α. Therefore, pharmacological targeting emerged as novel therapeutic approach recent years. The novelty this review it summarizes natural products colorectal not been presented earlier. We studied research publications related domain from 2010 2024. However, our main focus was identify better targeted for carcinoma management. Our described progression, employed inhibitors, discusses their potential inhibitors treatment.

Introduction Osteoarthritis (OA) is associated with excessive cartilage degradation, inflammation, and decreased autophagy. Insufficient efficacy of conventional monotherapies poor tissue regeneration due to side effects are just some the unresolved issues. Our previous research has shown that Calebin A (CA), a component turmeric ( Curcuma longa ), pronounced anti-inflammatory anti-oxidative by modulating various cell signaling pathways. Whether CA protects chondrocytes from degradation apoptosis in OA environment (EN), particularly via autophagy pathway, however completely unclear. Methods To study anti-degradative anti-apoptotic an inflamed joint, vitro model OA-EN was created treated antisense oligonucleotides targeting NF-κB (ASO-NF-κB), IκB kinase (IKK) inhibitor (BMS-345541) or 3-methyladenine (3-MA) and/or affect chondrocyte proliferation, apoptosis, The mechanisms underlying were investigated MTT assays, immunofluorescence, transmission electron microscopy, Western blot analysis 3D-OA high-density culture model. Results In contrast TNF-α-EN, treatment stress-induced defects inhibiting matrix pathways inflammation (NF-κB, MMP9) autophagy-repression (mTOR/PI3K/Akt), while promoting expression compounds (collagen II, specific proteoglycans), transcription factor Sox9, autophagy-associated proteins (Beclin-1, LC3). However, preventive properties could be partially abrogated 3-MA. Discussion present results reveal for first time able ameliorate progression chondrocytes, suggesting may novel therapeutic compound OA.
