Backgrounds: Melanogenesis, regulated by genetic, hormonal, and environmental factors, occurs in melanocytes the basal layer of epidermis. Dysregulation this process can lead to various skin disorders, such as hyperpigmentation hypopigmentation. Therefore, present study investigated effect ultrasonic-assisted ethanol extract (SHUE) from Sargassum horneri (S. horneri), brown seaweed against melanogenesis α-melanocyte-stimulating hormone (MSH)-stimulated B16F10 murine melanocytes. Methods: Firstly, yield proximate compositional analysis samples were conducted. The SHUE on cell viability has been evaluated using 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. After that, melanin content cellular tyrosinase activity α-MSH-stimulated examined. Western blot was carried out investigate protein expression levels microphthalmia-associated transcription factor (MITF), tyrosinase, tyrosinase-related protein-1 (TRP1), protein-2 (TRP2). In addition, extracellular signal-regulated kinase (ERK) assessed via blotting. Results: As per analysis, contained highest average a dry basis at 28.70 ± 3.21%. findings showed that reduced Additionally, MITF, TRP1, TRP2 significantly downregulated treatment Moreover, upregulated phosphorylation ERK AKT experiments conducted inhibitor (PD98059) revealed depends signaling cascade. Conclusion: These results suggest an anti-melanogenic be used material formulation cosmetics related whitening lightening.

Melasma is a prevalent chronic relapsing pigmentary disorder that affects photoexposed areas, especially in women of childbearing age. Although there currently no curative treatment available for melasma, this manuscript critically reviews the knowledge regarding photoprotection, topical and oral therapies, procedures such as peelings, laser, microneedling represent main strategies control prevention disease. As pathogenesis melasma not entirely understood, are prospects development new therapeutic might act on pathways promote sustained pigmentation rather than merely decreasing melanin synthesis removing from epidermis.

Arbutin, a hydroquinone glucoside, has been detected in ca. 50 plant families, especially the plants of Asteraceae, Ericaceae, Proteaceae and Rosaceae families. It is one most widely used natural skin-whitening agents. In addition to its skin whitening property, arbutin possesses other therapeutically relevant biological properties, e.g., antioxidant, antimicrobial anti-inflammatory, as well anticancer potential. This review presents, for first time, comprehensive overview distribution kingdom critically appraises therapeutic potential an agent based on literature published until end August 2022, accessed via several databases, Web Science, Science Direct, Dictionary Natural Products, PubMed Google Scholar. Excessive melanin deposition may lead to a series of skin disorders. The production is carried out by melanocytes, in which the enzyme tyrosinase performs key role. In this work, we identified novel inhibitor hybrids with dihydrochalcone skeleton and resorcinol structure, can inhibit activity reduce content skin. Compound 11c possessed most potent against tyrosinase, showing IC50 values at nanomolar concentration ranges, along significant antioxidant low cytotoxicity. Furthermore, vitro permeation tests, supported HPLC analysis 3D OrbiSIMS imaging visualization, revealed excellent 11c. More importantly, compound reduced on UV-induced pigmentation guinea pig model vivo. These results suggest that serve as promising for development potential therapy treat hyperpigmentation.
