Aflatoxin B1 is a toxin produced by the fungus Aspergillus flavus which reduces development and function of organs in broilers. The aim this study was to determine effect adding binder from different bentonite feed contaminated with AFB1 on productivity, intestinal morphology, liver toxicity A total 60-day old chick male broilers were placed 12 pens. Each treatment consisted three replicates, each replicate containing five Treatment P0 (control, basal diet, without addition AFB1), P1 (P0 + 100 µg/kg 4 g/kg calcium Type A), P2 B), P3 B kerolite saponite). diets given day 22 35 (finisher phase). results showed that had no consumption, body weight gain conversion (p>0.05). Aflatoxin B1 (AFB1) is the most dangerous and abundant mycotoxin, which toxic to almost all animals, poultry more sensitive AFB1 toxicity. Ingesting AFB1-contaminated feed can cause significant liver damage brings serious harm poultry, greatly restricts development of industry. The present research was implemented explore intervention effect its mechanism taraxasterol on induced by in broiler chickens. model chickens established feeding 0.5 mg/kg feed, (25, 50 100 BW, respectively) given drinking water for 21 days. growth performance, function, oxidative stress, apoptosis autophagy were evaluated. results showed that increased BW reduced feed-to-gain ratio AFB1. Taraxasterol improved levels serum alanine aminotransferase (ALT), aspartate (AST), γ-glutamyltransferase (GGT), total bilirubin (TBIL) alkaline phosphatase (ALP), attenuated hepatic histopathological changes Meantime, down-regulated cytochrome P450 (CYP450) enzyme system CYP1A1 CYP2A6 mRNA expression, inhibited overproduction reactive oxygen species (ROS) malondialdehyde (MDA), enhanced activities antioxidant enzymes glutathione (GSH) catalase (CAT) content superoxide dismutase (SOD) Furthermore, up-regulated protein expression nuclear factor E2 related 2 (Nrf2), heme oxygenase 1 (HO-1) NAD(P)H: quinone oxidoreductase (NQO1), kelch like ECH associated (Keap1) Keap1/Nrf2 signaling pathway. ultrastructural observation RT-qPCR found hepatocytes, B-cell lymphoma-2 (Bcl-2) Bax caspase3 mRNA. Further, restored hepatocytes phosphatidylinositol 3-kinase K (PI3K), kinase B (AKT) mammalian target rapamycin (mTOR) AFB1-induced above indicate alleviates through regulation pathway exert effect, mitochondrial improve anti-apoptotic ability PI3K/AKT/mTOR restore autophagy.

The gradual spread of Aspergilli worldwide is adding to the global shortage food and affecting its safe consumption. Aspergillus -derived mycotoxins, including aflatoxins ochratoxin A, fumonisins (members fusariotoxin group) can cause pathological damage vital organs, kidney or liver. Although functions as major excretory system in mammals, monitoring screening for mycotoxin induced nephrotoxicity only now a developmental area field livestock feed toxicology. Currently assessment individual exposure mycotoxins man animals usually based on analysis toxin and/or metabolite contamination blood urine. However, this requires selective sensitive analytical methods (e.g., HPLC-MS/MS), which are time consuming expensive. toxicokinetic metabolites becoming better understood. Several biomarkers used successfully drug development, however cost-efficient, reliable urgently needed farm early signs disease. β 2 -microglobulin (β -MG) N -acetyl-β-D-glucosaminidase (NAG) dominant employed routinely environmental toxicology research, while injury molecule 1 (KIM-1) neutrophil gelatinase-associated lipocalin (NGAL) also emerging effective markers identify nephropathy.
