Cu is an essential micronutrient for various physiological processes in almost all human cell types. Given the critical role of a wide range cellular processes, local concentrations and distribution transporter proteins lung are maintaining steady-state internal environment. Dysfunctional metabolism or regulatory pathways can lead to imbalance homeostasis lungs, affecting both acute chronic pathological processes. Recent studies have identified new form Cu-dependent death called cuproptosis, which has generated renewed interest diseases. Cuproptosis differs from other known pathways. This occurs through direct binding ions lipoylated components tricarboxylic acid cycle during mitochondrial respiration, leading aggregation subsequent downregulation Fe-S cluster proteins, causes toxic stress ultimately leads death. Here, we discuss impact dysregulated on pathogenesis respiratory diseases, including asthma, obstructive pulmonary disease, idiopathic interstitial fibrosis, cancer. We also therapeutic potential targeting Cu. study highlights intricate interplay between copper, health. Copper, while essential, must be carefully regulated maintain delicate balance necessity toxicity living organisms. review need further investigate precise mechanisms copper interactions with infections immune inflammation context diseases explore strategies related effects.

Abstract We present a novel lung aerosol exposure system named MALIES (modular air–liquid interface system), which allows three-dimensional cultivation of epithelial cells in alveolar-like scaffolds (MatriGrids ® ) and to nanoparticle aerosols. consists multiple modular units for generation, can be rapidly assembled commissioned. The complex process known as epithelial to mesenchymal transition (EMT) plays a fundamental role in several biological settings, encompassing embryonic development, wound healing, and pathological conditions such cancer fibrosis. In recent years, bulk of research has brought light the key copper, trace element with essential functions cellular metabolism, initiation progression. Indeed, besides functioning cofactor enzymes required for processes, energy production oxidation reactions, emerged an allosteric regulator kinases whose activity is fulfill dissemination through EMT. this comprehensive review, we try describe intricate relationship between metal copper EMT, spanning from earliest foundational studies latest advancements. Our aim shed on multifaceted roles undertaken by EMT unveil diverse mechanisms which homeostasis exerts its influence over regulators, signaling pathways, cell metabolic reprogramming transcription factors ultimately contributing spread cancer. Therefore, review not only may contribute deeper comprehension copper-mediated but also supports hypothesis that targeting counteract progression EMT-associated pathologies.

Copper (Cu) is an essential trace element for maintaining normal homeostasis in living organisms. Yet, elevated level of Cu beyond homeostatic capacity may lead to oxidative damage cellular components several organs, including the lungs. This work investigated effects curcumin (Curc) and nano-curcumin (nCurc) against Cu-induced lung injury, accenting roles stress, inflammation, nuclear factor erythroid 2-related factor/heme oxygenase-1 Nrf2/HO-1 pathway. Rats were challenged with 100 mg/kg copper sulfate (CuSO4) while being treated Curc or nCurc 7 days. Cu-triggered stress detected as dysregulation oxidative/antioxidant markers, a downregulation Nrf-2/HO-1 signaling, increase inflammatory markers interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α), intracellular adhesion molecule-1 (ICAM-1). Additionally, it decreased expression lung-specific proteins, surfactant protein-C (SP-C), mucin-1 (MUC-1), induced apoptosis, caused changes histology. alleviated CuSO4-induced injury by suppressing inflammation activating Nrf-2/HO-1. They also prevented apoptosis restored SP-C MUC-1. We concluded that exhibited superior efficacy compared mitigating injury. was associated reduced apoptotic responses increased signaling
