Cervical cancer remains a leading cause of cancer-related mortality in women low and middle-income countries despite efforts to improve prevention standard-of-care interventions. Sub-Saharan Africa (SSA) leads the numbers for global cervical incidence mortality, with majority diagnosed late stage malignancy. Although death rate has been on decline last two decades owing advancements screening treatment options, SSA not declined very much. Chemotherapy choice without meeting expected survival outcomes these patients, having advanced diseases at diagnosis. Immune checkpoint inhibitors have recently shown clinical promise improving patients integrated into guidelines most high-income countries, which helped further reduce cancer. Abstract: The utilization of PD-1/PD-L1 inhibitors marks a significant advancement in cancer therapy. However, the efficacy monotherapy is still disappointing substantial subset patients, necessitating exploration combinational strategies. Emerging from promising results KEYNOTE-942 trial, RNA-based therapies, particularly circRNAs and piRNAs, have distinguished themselves as innovative sensitizers to immune checkpoint (ICIs). These non-coding RNAs, notable for their stability specificity, were once underrecognized but are now known crucial roles regulating PD-L1 expression bolstering anti-cancer immunity. Our manuscript offers comprehensive analysis selected elucidating immunomodulatory effects mechanisms, thus underscoring potential ICIs enhancers. Colorectal cancer (CRC) initiating cells (CICs) possess self-renewal capabilities and are pivotal in tumor recurrence resistance to conventional therapies, including immunotherapy. The mechanisms underlying their interaction with immune remain unclear. We conducted a multi-omics analysis—encompassing DNA methylation, total RNA sequencing, microRNAs (miRNAs; N = 800) profiling on primary CICs differentiated cell lines, autologous pairs. Functional immunological assays were performed assess the impact of miRNA modulation. exhibited distinct methylation patterns, transcriptomic profiles, expressions compared (p < 0.05 or 0.01). Notably, miRNA-15a -196a implicated regulating tumorigenic pathways, such as epithelial-to-mesenchymal transition (EMT), TGF-β signaling, transfection mimics led downregulation oncogenic EMT markers (CRKL, lncRNA SOX2-OT, JUNB, SMAD3) pathway, resulting significant reduction vitro proliferation tumorigenicity migration zebrafish xenograft model. Additionally, enhanced expression antigen processing machinery decreased checkpoints (PD-L1, PD-L2, CTLA-4) immunosuppressive cytokines (IL-4). co-culture HLA-matched lymphocytes overexpressing miRNA-15a, elicited robust tumor-specific responses, characterized by shift toward central effector memory T phenotypes prevented terminal differentiation exhaustion. combination modulation Indoleamine 2,3-dioxygenase blockade immunomodulating agents further potentiated these effects. Abstract Background and Objective A disintegrin metalloprotease 33 (ADAM33) is associated with asthma susceptibility, its genetic variations impact susceptibility disease severity. However, the mechanisms remain unclear. This study aimed to investigate ADAM33 single nucleotide polymorphisms (SNPs) in childhood explore their regulatory mechanisms. Methods Eleven selected SNPs were genotyped identified association susceptibility. In validation cohort, we measured plasma sADAM33 levels compared them severity among children different SNP genotypes. Computational predictions miRNAs targeting SNP, of on miRNA regulation was confirmed using a dual luciferase reporter system. Finally, validated role expression an vitro model upregulated expression. Results Only rs3918400 allergic exhibited higher levels.
