In mammalian cells, the content of polyamines is tightly regulated. Polyamines, including spermine, spermidine and putrescine, are involved in many cellular processes. Spermine oxidase specifically oxidizes its deregulated activity has been reported to be linked brain pathologies involving neuron damage. a neuromodulator number ionotropic glutamate receptors types ion channels. this respect, Dach-SMOX mouse model overexpressing spermine neocortex neurons was revealed chronic oxidative stress, excitotoxicity neuronal Reactive astrocytosis, excitotoxic loss susceptibility seizure discussed here. This genetic would help researchers understand linkage between polyamine dysregulation neurodegeneration unveil roles crosstalk astrocytes neuroprotection or neurodegeneration.

By replacing a phenolic ring of (E)-resveratrol with an 1,3,4-oxadiazol-2(3H)-one heterocycle, new resveratrol-based multitarget-directed ligands (MTDLs) were obtained. They evaluated in several assays related to oxidative stress and inflammation (monoamine oxidases, nuclear erythroid 2-related factor, quinone reductase-2, oxygen radical trapping) then experiments increasing complexity (neurogenic properties neuroprotection vs okadaic acid). 5-[(E)-2-(4-Methoxyphenyl)ethenyl]-3-(prop-2-yn-1-yl)-1,3,4-oxadiazol-2(3H)-one (4e) showed well-balanced MTDL profile: cellular activation the NRF2-ARE pathway (CD = 9.83 μM), selective inhibition both hMAO-B QR2 (IC50s 8.05 0.57 best ability promote hippocampal neurogenesis. It good drug-like profile (positive vitro central nervous system permeability, physiological solubility, no glutathione conjugation, lack PAINS or Lipinski alerts) exerted neuroprotective antioxidant actions acute chronic Alzheimer models using tissues. Thus, 4e is interesting that could stimulate defensive regenerative pathways block early events neurodegenerative cascades.

Intracerebroventricularly (icv) injected streptozotocin (STZ) is a widely used model for sporadic Alzheimer's disease (sAD)-like pathology, marked by oxidative stress-mediated pathological progression. Intermittent theta burst stimulation (iTBS) noninvasive technique brain activity with the ability to induce long-term potentiation-like plasticity and represents promising treatment several neurological diseases, including AD. The present study aims investigate effect of iTBS protocol on animal STZ-induced sAD-like pathology in context antioxidant, anti-inflammatory, anti-amyloidogenic effects cortex, striatum, hippocampus, cerebellum.Male Wistar rats were divided into four experimental groups: control (icv normal saline solution), STZ STZ-3 mg/kg), + (STZ subjected protocol), Placebo animals placebo noise artifact). Biochemical assays immunofluorescence microscopy evaluate functional structural changes. The icv administration induces stress attenuates antioxidative capacity all examined regions. significantly reduced nitrosative parameters. Also, decreased Aβ-1-42 APP levels. enhances reported as elevated its enzymatic non-enzymatic components. In addition, BDNF expression attenuated astrogliosis confirmed GFAP+/VIM+/C3+ cell reactivity hippocampus. Our results provide evidence beneficial applied attenuating stress, increasing antioxidant decreasing reactive STZ-administrated rats.
