Calcipotriol Attenuates Form Deprivation Myopia Through a Signaling Pathway Parallel to TGF-β2–Induced Increases in Collagen Expression DOI Creative Commons

Purpose: To determine the role of calcipotriol, a vitamin D3 analogue, in myopia development and altering expression scleral α1 chain type I collagen (Col1α1) mice. We also aimed to identify if signaling pathway mediating above changes is different from one involved transforming growth factor β2 (TGF-β2)–mediated increases COL1A1 cultured human fibroblasts (HSFs). Methods: C57BL/6J mice were either intraperitoneally injected with calcipotriol subjected form deprivation (FD) or exposed normal refractive for 4 weeks. Scleral D receptor (Vdr) was knocked down using Sub-Tenon's capsule injection an adeno-associated virus–packaged short hairpin RNA (AAV8-shRNA). Refraction biometric measurements evaluated development. A combination knockdown induction strategies determined relative contributions TGF-β2 pathways modulating HSFs. Results: Calcipotriol injections suppressed FD-induced (FDM), but it had no significant effect on AAV8-shRNA reduced Vdr mRNA by 42% shifted refraction toward (−3.15 ± 0.99D, means SEM) eyes. In HSFs, VDR calcipotriol-induced rises expression, did not alter TGF-β2–induced expression. Additionally, augmented TGF-β (TGFBRI/II) blunted whereas levels unaltered. Conclusions: inhibits mice, potentially activating VDR-dependent increasing levels.
