Cryopreservation of gametes is one the important stages assisted reproductive technologies (ART). To obtain high results using ART, it essential to cryopreserve sperm in a favorable season, when have ability fertilize oocytes. The aim this research was study seasonal variability spermatozoa cryoresistance and characteristics Saanen goats. After removing seminal fluid, five mature animals were cryopreserved solution 10% glycerol 20% egg yolk. examined by embryo development rate on day 7 vitro culture. Spermatozoa need to undergo an exocytotic event called the acrosome reaction before fusing with eggs. Although calcium ion (Ca 2+ ) is essential for reaction, its molecular mechanism remains unknown. Ferlin a single transmembrane protein multiple Ca -binding C2 domains, and there are six ferlins, dysferlin (DYSF), otoferlin (OTOF), myoferlin (MYOF), fer-1–like 4 (FER1L4), FER1L5, FER1L6, in mammals. Dysf , Otof Myof knockout mice have been generated, each mouse line exhibited membrane fusion disorders such as muscular dystrophy deafness abnormal myogenesis . Here, by generating mutant of Fer1l4 Fer1l5 Fer1l6 we found that only required male fertility. spermatozoa could migrate female reproductive tract reach eggs, but no took place. Since artificial insemination is common practice in pig breeding, the quality and persistence of semen are decisive for usability individual boars. In current study, genome-wide association analyses were performed to investigate genetic variability underlying phenotypic variations characteristics. To improve artificial insemination protocols in ovine species it is crucial to optimize sperm quality evaluation after preservation technologies. Sperm cells are of unique elongated structure and function, the development which is tightly regulated by existing proteins posttranslational modifications (PTM) these proteins. Based on phylogenetic relationships various swine breeds, Yorkshire boar believed to be distinctly different from Duroc boar. The comprehensive differential proteomics phosphoproteomics profilings were performed spermatozoa both boars. By peptide PTM quantification followed statistical analyses, 167 differentially expressed identified 1,745 proteins, 283 phosphopeptides corresponding 102 phosphorylated measured 1,140 derived 363 representative results validated Western blots. Pathway enrichment analyses revealed that majority expression phosphorylation primarily concerned with spermatogenesis, male gamete generation, sperm motility, energy metabolism, cilium morphogenesis, axonemal dynein complex assembly, sperm–egg recognition, capacitation.
