ATP-citrate lyase (ACLY) generates cytosolic acetyl-coenzyme A (acetyl-CoA) for lipid synthesis and is a promising therapeutic target in diseases with altered metabolism. Here, we developed inducible whole-body Acly-knockout mice to determine the requirement ACLY normal tissue functions, uncovering its crucial role skin homeostasis. ACLY-deficient upregulates acetyl-CoA synthetase ACSS2; deletion of both Acly Acss2 from exacerbates abnormalities, differential effects on two major lipid-producing compartments. The investigation into promising botanical materials for natural cosmetics is expanding due to environmental and health awareness. Here, we aimed evaluate the phytochemical substances potential skin-related pharmacological activities of four Mucuna seeds, namely M. gigantea (Willd.) DC. (MGG), interrupta Gagnep. (MIT), monosperma Wight (MMM), pruriens (L.) (MPR), belonging Fabaceae family. In methodology, seeds were authenticated using morphological molecular approaches. L-DOPA, phenolics, flavonoid content, incorporated with HPLC GC–MS fingerprinting analyses, determined. Then, antimicrobial, antioxidant, antiaging results revealed that MPR showed highest L-DOPA content (75.94 mg/100 mg extract), whereas MGG exhibited phenolic (56.73 ± 0.62 gallic/g extract 1030.11 3.97 quercetin/g extract, respectively). Only MMM could inhibit all S. aureus, epidermidis, C. albicans, but no sample acnes. Furthermore, samples demonstrated antioxidant activity. Interestingly, strong collagenase, elastase, hyaluronidase inhibitory activities. We conclude ethanolic extracts are probably advantageous in development skincare cosmeceutical products.
