During the progression of knee osteoarthritis (OA), synovium and infrapatellar fat pad (IFP) can serve as source for Substance P (SP) calcitonin gene-related peptide (CGRP), two important pain-transmitting, immune, inflammation modulating neuropeptides. Our previous studies showed that pad-derived mesenchymal stem/stromal cells (MSC) acquire a potent immunomodulatory phenotype actively degrade via CD10 both in vitro vivo. On this basis, our hypothesis is CD10-bound IFP-MSC sEVs be engineered to target CGRP while retaining their anti-inflammatory phenotype. Herein, human cultures were transduced with an adeno-associated virus (AAV) vector carrying GFP-labelled gene antagonist (aCGRP). The GFP positive aCGRP isolated sEVs’ miRNA protein cargos assessed using multiplex methods. results purified yielded cargo 147 distinct MSC-related miRNAs. Reactome analysis miRNAs detected these revealed strong involvement regulation genes involved pathways control pain, cartilage homeostasis. Protein array demonstrated high presence key reparative proteins. Stimulated macrophages exposed switch towards alternate M2 status. Also, stimulated cortical neurons modulate molecular pain signaling profile. Collectively, data suggest putatively vivo, containing analgesic cargo, suggesting promise novel sEVs-based therapeutic approaches diseases such OA.

Circular RNAs (CircRNAs) are reported to exert vital regulatory roles in the occurrence and development of various human malignancies, including non-small cell lung cancer (NSCLC). Bioinformatics methods identified down-regulation circSCN8A (circBase ID: hsa_circ_0026337) NSCLC tissues. However, its biological functions molecular mechanisms remain unknown. In this study, we found that expression was down-regulated tissues cells. Low positively associated with aggressive clinicopathological characteristics poor prognosis patients. CircSCN8A suppressed proliferation, migration, invasion, epithelial-mesenchymal transition (EMT) vitro blocked tumor growth vivo. Moreover, promoted ferroptosis NSCLC. Mechanistically, acted as a competing endogenous RNA (ceRNA) by sponging miR-1290 enhance long-chain acyl-CoA synthetase-4 (ACSL4). Furthermore, knockdown ACSL4 or overexpression reversed effect on facilitating inhibiting proliferation metastasis. summary, represses metastasis regulating miR-1290/ACSL4 axis induce ferroptosis. Thus, may represent promising therapeutic target against

Epithelial ovarian cancer (EOC) remains a leading cause of gynecologic mortality. Despite advances in treatment, metastatic progression and resistance to standard therapies significantly worsen patient outcomes. Epithelial–mesenchymal transition (EMT) is critical process metastasis, enabling cells gain invasive migratory capabilities, often driven by changing miRNA expression involved the regulation pathological processes like drug resistance. Targeted PARP inhibitors (PARPi) have improved outcomes, particularly BRCA-mutated DNA repair-deficient tumors; however, limited efficacy advanced stages remain challenges. Recent studies highlight potential synergy PARPi with damage response (DDR) inhibitors, such as ATR CHK1 which disrupt cell survival pathways under stress. This study investigated combined effects olaparib (ATRi CHK1i) on migration, invasion, EMT-related protein lines OV-90 SKOV-3. The results demonstrated enhanced cytotoxicity, inhibition migration modulation miRNAs linked metastasis. These findings suggest that combination targeting repair cycle may offer novel, more effective approach managing EOC reducing spread.

microRNAs are regulatory RNAs that silence specific mRNA by binding to it, inducing translational repression. Over the recent decades since discovery of RNA interference, field microRNA therapeutics has expanded tremendously. The role miRNAs in disease development attracted researchers investigate their potential therapeutics. In lung cancer, multiple deregulated, and involvement is observed cell proliferation, immunomodulation, angiogenesis, epithelial-mesenchymal transition. Thus, synthetic oligonucleotides developed downregulate overexpressed miRNA or upregulate repressed miRNA. However, clinical efficiency limited due requirement for an effective delivery strategy. Advances current understanding nanotechnology, biomaterial science, molecular pathology have increased chances overcoming limitations miRNA-based therapy. The purpose our study is demonstrate precision a panel miRNAs with potential for risk stratification. present demonstrates comparative expression profiles miRNA-141,-1290,-100 and-335 both tissue serum including Benign Prostate Hyperplasia (BPH) PCa healthy volunteers. Firstly, we all discovery cohort metastasis benign later validate their non-invasive MiRNA was isolated from be quantified by RT-PCR analyzed biomarker ROC curve analysis followed targetome each miRNA. Among miRNA assessed, it seen that 141 (p=0.0003) 1290 (p< 0.0001) are oncogenic significantly higher while 100 (p=0.0002) 335 tumor suppressor PCa, compared controls. While BPH, (p=0.003) were found suppressor, respectively. (miRNA-141,-1290, -100) portrayed significant area under greater sensitivity specificity.
