Abstract Dietary fiber plays a significant role in gastrointestinal health. In order to thoroughly comprehend the protective effect of dietary on calf intestinal health, we conducted randomized controlled trial with135 calves. We profiled gut bacterial community fecal samples from calves without diarrhea at five time points (0, 7, 14, 28, and 56 days age) using both 16s amplicon sequencing metagenomic sequencing. Our findings revealed significantly elevated abundance Bifidobacterium Prevotella intestine fed with compared control group. may mitigate damage protect health by modulating secondary bile acid metabolism flavonoid pathways. Furthermore, CAZy differential analysis reveals that feeding different fibers can induce increased CAZymes. also found changes CAZymes induced are associated distinct microbial communities. This suggests stimulates colonization species intestine, thereby increasing activity carbohydrate enzymes microbiota. lays foundation for management during transition period. Additionally, our research psyllium husk powder age harbored intergeneric symbiotic clusters their intestines consisting Clostridium, Prevotella, Bacteriodes, others. The human gut microbiota, an intricate ecosystem within the gastrointestinal tract, plays a pivotal role in health and disease. Prebiotics, non-digestible food ingredients that beneficially affect host by selectively stimulating growth and/or activity of beneficial microorganisms, have emerged as key modulator this complex microbial community. This review article explores evolution prebiotic concept, delineates various types prebiotics, including fructans, galactooligosaccharides, xylooligosaccharides, chitooligosaccharides, lactulose, resistant starch, polyphenols, elucidates their impact on microbiota composition. We delve into mechanisms through which prebiotics exert effects, particularly focusing producing short-chain fatty acids modulating towards health-promoting implications are extensively reviewed, conditions such obesity, inflammatory bowel disease, immune function, mental health. further discusses emerging concept synbiotics—combinations probiotics synergistically enhance health—and highlights market potential response to growing demand for functional foods. By consolidating current knowledge identifying areas future research, aims understanding prebiotics’ underscoring importance maintaining healthy microbiome overall well-being.

Inflammatory bowel disease (IBD), including Crohn's and ulcerative colitis, is a complex disorder with an unknown cause. However, the dysbiosis of gut microbiome has been found to play role in IBD etiology, exacerbated immune responses defective intestinal barrier integrity. The can also be potential biomarker for several diseases, IBD. Currently, conventional treatments targeting pro-inflammatory cytokines pathways IBD-associated do not yield effective results. Other therapies that directly target dysbiotic outcomes are emerging. We review health its as diagnostic, prognostic, therapeutic This explores emerging advancements microbiome-associated alterations IBD, such nanoparticle or encapsulation delivery, fecal microbiota transplantation, nutritional therapies, microbiome/probiotic engineering, phage therapy, mesenchymal stem cells (MSCs), proteins, herbal formulas.

Circadian rhythm plays an important role in intestinal homeostasis and immune function. dysregulation was reported to induce microbiota dysbiosis, barrier disruption, trigger inflammation. However, the relationship between metabolites circadian of still unclear. Urolithin A (UA), a kind microbial metabolite, selected this study. Results showed UA influenced on expression clock genes BMAL1 PER2 epithelial cells. Furthermore, study investigated effects rhythms (BMAL1 PER2) tight junctions (OCLN, TJP1, CLND1), all which were dysregulated by In addition, pre-treatment oral administration female C57BL/6 mice improvement fecal IgA concentrations, junction (Clnd1 Clnd4), gene (Bmal1 Per2) DSS-induced colitis model induced using DSS treatment. Finally, Nrf2-SIRT1 signaling pathway confirmed be involved UA’s effect cells antagonist This also evidence that feeding impact central clock, are SCN. Abstract In the current study, we utilized molecular modeling and simulation approaches to define putative potential targets for Burdock Inulin, including inflammatory proteins such as iNOS, COX-2, TNF-alpha, IL-6, IL-1β. Molecular docking results revealed interactions good binding affinity these targets; however, IL-1β, iNOS were identified best Inulin. simulation-based stability assessment demonstrated that inulin could primarily target may also supplementarily COX-2 IL-1β during DSS-induced colitis reduce role of mechanisms. Furthermore, residual flexibility, hydrogen bonding, structural packing reported with uniform trajectories, showing no significant perturbation throughout simulation. The protein motions within trajectories clustered using principal component analysis (PCA). IL-1β–Inulin complex, approximately 70% total motion was attributed first three eigenvectors, while remaining contributed by eigenvectors. contrast, COX2–Inulin 75% in iNOS–Inulin eigenvectors 60% motion. complex through To explore thermodynamically favorable changes upon mutation, mode carried out. Free Energy Landscape (FEL) achieved a single conformation lowest energy, exhibited two lowest-energy conformations each. displayed free energies − 27.76 kcal/mol 37.78 kcal/mol, respectively, energy at 45.89 kcal/mol. This indicates stronger pharmacological than other complexes. Thus, further experiments are needed use autoimmune diseases.
