Background/Objectives: Côa Valley, located in the northeast of Portugal, harbors more than 500 medicinal plant species. Among them, four species stand out due to their traditional uses: Equisetum ramosissimum Desf. (hemorrhages, urethritis, hepatitis), Rumex scutatus L. subsp. induratus (Boiss. and Reut.) Malag. (inflammation, constipation), Geranium purpureum Vill., lucidum (pain relief, gastric issues). Given rich ethnomedicinal history, we evaluated protective effects on an vitro model metabolic dysfunction-associated steatotic liver disease (MASLD). Methods: Decoction (D) hydroalcoholic (EtOH80%) extracts were prepared chemically characterized. Their safety profile lipid accumulation assessed palmitic acid (PA)-treated HepG2 cells using resazurin, sulforhodamine B, Nile Red assays. Results: Chemical analysis revealed diverse phenolic compounds, particularly kaempferol derivatives E. ramosissimum. All showed minimal cytotoxicity at 25–50 µg/mL. At 100 µg/mL, only maintained high cell viability. In lipotoxicity model, decoction demonstrated most potent effect, significantly reducing PA-induced neutral a dose-dependent manner, while other varying degrees activity. Conclusions: These findings highlight ramosissimum’s decoction, derivatives, as effective this MASLD also providing comprehensive characterization traditionally used plants from Valley region.

Non-alcoholic fatty liver disease (NAFLD) is one of the most common diseases. Its incidence progressively rising and it possibly becoming a worldwide epidemic. NAFLD encompasses spectrum diseases accounting for chronic accumulation fat within hepatocytes due to various causes, excluding excessive alcohol consumption. In this study, we aimed focus on finding evidence regarding implications oxidative stress inflammatory processes that form multifaceted pathophysiological tableau in relation thrombotic events co-occur associated Recent pathophysiology suggests complex pattern multidirectional components, such as prooxidative, proinflammatory, prothrombotic better explains multiple factors promote mechanisms underlying acid excess subsequent processes. As there extensive multi-component nature pathophysiology, further studies could address interactions underlie development progression disease. Therefore, study describe possible connecting molecular impairments with clinical manifestations, focusing especially among stress, inflammation, coagulation dysfunctions. Thus, described bidirectional modulation homeostasis, inflammation occurs stages NAFLD.

The aqueous extract of mulberry (MLE) various biologically active components such polyphenols and flavonoids. However, inhibitory effect MLE in hepatocarcinogenesis is poorly understood. In this study, we determined role supplementation preventing carcinogen-initiated high-fat diet (HFD)-promoted Sprague-Dawley (SD) rat model. rats were fed an HFD to induce obesity spontaneous hepatomas by administering 0.01% diethylnitrosamine (DEN) their drinking water for 12 weeks (HD group), also through oral ingestion at daily doses 0.5%, 1%, or 2%. At end 12-week experimental period, liver tumors analyzed identify markers oxidative stress antioxidant enzyme activities, serum determine nutritional status function. Histopathological analysis revealed that significantly suppressed severity incidence hepatic tumors. Furthermore, compared with + DEN groups, expression protein kinase C (PKC)-α Rac family small GTPase 1 (Rac1) lower groups. These findings suggest prevents obesity-enhanced, carcinogen-induced hepatocellular carcinoma development, potentially (PKC)α/Rac1 signaling pathway. might be effective chemoprevention modality nonalcoholic fatty disease (NAFLD)/nonalcoholic steatohepatitis (NASH)-related hepatocarcinogenesis.

Non-alcoholic fatty liver disease (NAFLD) is the most prevalent form of disease. Orthosiphon aristatus (Blume) Miq, a traditional plant in South Asia, has previously been shown to attenuate obesity and hyperglycaemic conditions. Eight weeks feeding C57BL/6 mice with standardized O. extract (400 mg/kg) inhibited progression NAFLD. Liver enzymes including alanine aminotransferase aspartate transaminase were significantly reduced treated by 74.2% ± 7.69 52.8% 7.83, respectively. Furthermore, showed reduction serum levels glucose (50% 5.71), insulin (70.2% 12.09), total cholesterol (27.5% 15.93), triglycerides (63.2% 16.5), low-density lipoprotein (62.5% 4.93) atherogenic risk index relative negative control. Histologically, reversed hepatic fat accumulation NAFLD severity. Notably, our results antioxidant activity via increased superoxide dismutase malondialdehyde levels. In addition, pro-inflammatory mediators (IL-6 TNFα) decreased, indicating anti-inflammatory activity. The aqueous, hydroethanolic ethanolic fractions intracellular HepG2 cells that palmitic-oleic acid. Together, these findings suggest activities are primary mechanism action underlying anti-NAFLD effects.
