Aging is a multifaceted process influenced by hereditary factors, lifestyle, and environmental elements. As time progresses, the human body experiences degenerative changes in major functions. The external internal signs of aging manifest various ways, including skin dryness, wrinkles, musculoskeletal disorders, cardiovascular diseases, diabetes, neurodegenerative cancer. Additionally, cancer, like aging, complex disease that arises from accumulation genetic epigenetic alterations. Circadian clock dysregulation has recently been identified as an important risk factor for cancer development. Natural compounds herbal medicines have gained significant attention their potential preventing age-related diseases inhibiting progression. These demonstrate antioxidant, anti-inflammatory, anti-proliferative, pro-apoptotic, anti-metastatic, anti-angiogenic effects well circadian regulation. This review explores cancers, specific natural targeting key features these conditions.

Chondrocyte apoptosis is associated with the severity of cartilage destruction and matrix degeneration in progression osteoarthritis. Increasing evidence indicates that autophagy has a significant cytoprotective effect against chondrocyte apoptosis. Here, we investigated role microRNA-103-3p (miR-103-3p) regulating function elucidated underlying mechanism. MiR-103-3p expression interleukin-1β (IL-1β)-stimulated chondrocytes was evaluated using RT-qPCR. The targets miR-103-3p predicted by online databases were verified biotin-based pulldown assay luciferase reporter assay. IL-1β stimulated-chondrocytes transfected inhibitor along siRNA targeting cytoplasmic polyadenylation element-binding protein3 (siCPEB3), 3-MA, or PI3K agonist 740 Y-P. proliferation cell counting kit-8. Apoptosis detected flow cytometry. levels apoptosis-, extracellular (ECM)-, autophagy-, PI3K/Akt/mTOR pathway-related proteins immunoblotting immunofluorescence. We found stimulation upregulated downregulated CPEB3 mouse chondrocytes. Inhibiting reduced IL-1β-induced ECM macromolecule degradation while enhancing targeted CPEB3, its downregulation rescued level stimulated-chondrocytes. inhibited pathway upregulating CPEB3. Y-P, knockdown counteracted on apoptosis, degradation, autophagy. Overall, inhibition reduces through CPEB3/PI3K/Akt/mTOR pathway.

Osteoarthritis (OA) is a disease of the joints, characterized by chronic inflammation, cartilage destruction and extracellular matrix (ECM) remodeling. Aberrant chondrocyte hypertrophy promotes OA development. Collagen X, biomarker hypertrophy, upregulated runt‑related transcription factor 2 (Runx2), which mediated bone morphogenetic protein 4 (BMP4)/Smad1 signaling pathway. BMP binding endothelial regulator (BMPER), secreted glycoprotein, acts as an agonist BMP4. 5,7,3',4'‑tetramethoxyflavone (TMF) natural flavonoid derived from Murraya exotica L. Results our previous study demonstrated that TMF exhibits chondroprotective effects against development through activation Forkhead box O3a (FOXO3a) expression. However, whether suppresses FOXO3a expression inhibition BMPER/BMP4/Smad1 remains unknown. present revealed inhibited collagen X Runx2 expression, signaling, activated expression; thus, protecting BMPER overexpression knockdown impacted protective TMF. Thus, in mediating FOXO3a/BMPER

Introduction : Osteoarthritis is a prevalent degenerative joint disorder associated with pain and functional impairment. Curcumin, natural anti-inflammatory compound, has garnered attention for its potential therapeutic benefits in osteoarthritis management. Aim This study aimed to investigate the correlations between curcumin use, patient characteristics, medication changes health outcomes among patients. The primary hypothesis was that use would correlate positive metrics. Materials methods A cohort of 60 patients enrolled this observational divided into 4 groups according type osteoarthritis. Patient selection criteria included diagnosis consent supplementation. outcome variable metrics measured across three visits. Body mass index, nonsteroidal drugs (NSAIDs) paracetamol were also recorded. Correlation analysis paired t -tests employed examine relationships variables Results analyses revealed weak negative correlation BMI ‘Visit 1’ 2’ ( r =−0.09). NSAIDs exhibited during period =0.17), while displayed =−0.05) changes. Statistically significant observed (mean change =−5.93, p <0.05), aligning prior studies indicating curcumin’s symptom relief. Conclusions provides valuable insights intricate exist curcumin, Despite many limitations, including sample size research duration, our indicates may have impact on symptomatology findings highlight need more studies, larger trials, mechanistic completely understand role

Degenerative bone and joint diseases (DBJDs), characterized by osteoporosis, osteoarthritis, chronic inflammation of surrounding soft tissues, are systemic conditions primarily affecting the skeletal system. Ferroptosis, a programmed cell death pathway distinct from apoptosis, autophagy, necroptosis. Accumulating evidence suggests that ferroptosis is intricately linked to pathogenesis DBJDs, targeting its regulation could be beneficial in managing these conditions. Natural products, known for their anti-inflammatory antioxidant properties, have shown unique advantages preventing potentially through modulating ferroptosis. This article provides an overview latest research on ferroptosis, with focus role DBJDs therapeutic potential natural products this pathway, offering novel insights prevention treatment DBJDs.
