Nanotechnology is an interdisciplinary domain of science, technology and engineering that deals with nano-sized materials/particles. Usually, the size nanoparticles lies between 1 100 nm. Due to their small large surface area-to-volume ratio, exhibit high reactivity, greater stability adsorption capacity. These important physicochemical properties attract scientific community utilize them in biomedical field. Various types (inorganic organic) have broad applications medical field ranging from imaging gene therapy. are also effective drug carriers. In recent times, utilized circumvent different treatment limitations. For example, ability cross blood-brain barrier having a certain degree specificity towards amyloid deposits makes themselves candidates for Alzheimer's disease. Furthermore, nanotechnology has been used extensively overcome several pertinent issues like drug-resistance phenomenon, side effects conventional drugs targeted delivery issue leprosy, tuberculosis cancer. Thus, this review, application these four diseases (Alzheimer's disease, tuberculosis, leprosy cancer) as well presented systematically. Although nanoformulations many advantages over traditional therapeutics treating diseases, nanotoxicity major concern discussed subsequently. Lastly, we promising future prospective alternative therapeutics. section, about futuristic approach(es) could provide candidate(s) diseases.

Abstract Alzheimer's disease (AD) is the most common age‐related neurodegenerative disorder leading to cognitive decline. Excessive cytosolic calcium (Ca 2+ ) accumulation plays a critical role in pathogenesis of AD since it activates NOD‐like receptor family, pyrin domain containing 3 (NLRP3), switches endoplasmic reticulum (ER) unfolded protein response (UPR) toward proapoptotic signaling and promotes A β seeding. Herein, liposomal nanodrug (felodipine@LND) developed incorporating channel antagonist felodipine for treatment through low‐intensity pulse ultrasound (LIPUS) irradiation‐assisted blood brain barrier (BBB)‐crossing drug delivery. The multifunctional felodipine@LND effectively delivered diseased applying LIPUS irradiation skull, which resulted series positive effects against AD. Markedly, switched ER UPR antioxidant signaling, prevented surface translocation calreticulin (CALR) microglia, inhibited NLRP3 activation In addition, promoted degradation damaged mitochondria via mitophagy, thereby inhibiting neuronal apoptosis. Therefore, anxiety‐like behavior impairment 5xFAD mice with significantly ameliorated, manifested potential – assisted BBB‐crossing delivery serve as paradigm therapy based on well‐recognized clinically available felodipine.

Abstract Neurodegenerative diseases (NDs) impose substantial medical and public health burdens on people worldwide represent one of the major threats to human health. The prevalence these age‐dependent disorders is dramatically increasing over time, a process intrinsically related constantly rising percentage elderly population in recent years. Among all NDs, Alzheimer's Parkinson's are considered most debilitating as they cause memory cognitive loss, well severely affecting basic physiological conditions such ability move, speak, breathe. There an extreme need for new more effective therapies counteract devastating diseases, available treatments only able slow down pathogenic without really stopping or resolving it. This review aims elucidate current nanotechnology‐based tools representing future hope NDs treatment. Noble metal nano‐systems, that is, gold silver nanoparticles (NPs), have indeed unique physicochemical characteristics enabling them deliver any pharmacological treatment way within central nervous system. can potentially make NPs reversing actual therapeutic strategy based slowing irreversible into permanent
