Patients suffering from chronic pain often present with multifactorial underlying conditions, sometimes without concrete pathological physical findings. Functional somatic syndromes (FSS) and somatoform disorders show a high prevalence of 8-20% are associated adverse childhood experiences (ACE) stress. As many different FSS have overlapping symptoms, the concept multisomatoform disorder (MSD) has been introduced as an encompassing concept. We hypothesize that common neurohumoral profile is in patients MSD distinct gender- age-matched controls thus provides insight into possible mechanisms. In 151 (138 females) 149 matched (131 females), we determined ACE by Childhood Trauma Questionnaire (CTQ) stress Trier Inventory for Chronic Stress (TICS). Furthermore, serum levels leptin, FSH, LH, cortisol, DHEA-S, IGF-1 assessed. There were significant differences IGF-1, cortisol between controls, mainly driven female participants. Levels leptin significantly correlated BMI patients, subgroup. This correlation was exaggerated when compared to controls. Both CTQ TICS predicted directly indirectly through leptin. There evidence healthy similar what demonstrated other states. The endocannabinoid system (ECS) is a complex cell-signaling that responsible for maintaining homeostasis by modulating various regulatory reactions in response to internal and environmental changes. influence of ECS on appetite regulation has been subject much recent research, however, the full extent its impact remains unknown. Current evidence links human obesity activation, increased levels both central peripheral tissues, along with cannabinoid receptor type 1 (CBR1) up-regulation. These findings imply potential pharmacological use treatment obesity. Here, we present pathophysiological processes involving ECS, highlighting different options activity treat However, those possibilities under investigation requires further research.

Abstract Associations between brain and obesity are bidirectional: changes in structure function underpin over-eating, while chronic adiposity leads to atrophy. Investigating brain-obesity interactions across the lifespan can help better understand these relationships. This study explores interaction cortical morphometry children, young adults, older adults. We also investigate genetic, neurochemical, cognitive correlates of associations. Our findings reveal a pattern lower thickness fronto-temporal regions associated with all age cohorts varying age-dependent patterns remaining regions. In adults neurochemical expression inflammatory mitochondrial genes. children is modifications involved emotional attentional processes. Thus, might originate from during early adolescence, leading neurodegeneration later life through mechanisms.

Inflammatory bowel disease (IBD) is a general term used to describe group of chronic inflammatory conditions the gastrointestinal tract unknown etiology, including two primary forms: Crohn's (CD) and ulcerative colitis (UC). The endocannabinoid system (ECS) plays an important role in modulating many physiological processes intestinal homeostasis, modulation motility, visceral sensation, or immunomodulation inflammation IBD. It consists cannabinoid receptors (CB1 CB2), transporters for cellular uptake ligands, endogenous bioactive lipids (Anandamide 2-arachidonoylglycerol), enzymes responsible their synthesis degradation (fatty acid amide hydrolase monoacylglycerol lipase), manipulation which through agonists antagonists system, shows potential therapeutic ECS disease. Cannabis sativa is known for its therapeutic benefit in various diseases including pain relief by targeting cannabinoid receptors. The primary component of cannabis, Δ9-tetrahydrocannabinol (THC), and other agonists engage the orthosteric site CB1, activating both Gi β-arrestin signaling pathways. activation diverse pathways could result on-target side effects cannabis addiction, which may hinder potential. A significant challenge pharmacology design a ligand that can modulate specific CB1. By leveraging insights from structure–function selectivity relationship (SFSR), we have identified signaling–biased agonist-allosteric modulators (ago-BAMs). Further, two cryoelectron microscopy (cryo-EM) structures reveal binding mode ago-BAM at extrahelical allosteric Combining mutagenesis pharmacological studies, elucidated detailed mechanism ago-BAM-mediated biased signaling. Notably, CB-05 demonstrated analgesic efficacy with fewer effects, minimal drug toxicity no addiction mouse models. In summary, our finding not only suggests ago-BAMs CB1 provide potential nonopioid strategy management but also sheds light on BAM identification GPCRs.
