Abstract Increasing evidence has shown that many environmental and toxic factors can cause testicular damage, leading to ferroptosis subsequent male reproductive disorders. Melatonin is a major hormone plays an vital role in regulating reproduction. However, there lack of research on whether Mel alleviate cell its specific mechanism. In this study, the results indicated could enhance viability swine testis cells undergoing ferroptosis, reduce LDH enzyme release, increase mitochondrial membrane potential, affect expression biomarkers. Furthermore, we found melatonin depended receptor 1B exert these functions. Detection MMP biomarker protein confirmed MT2 acted through downstream Akt signaling pathway. Moreover, inhibition pathway eliminate protective effect inhibit AMPK phosphorylation, gated channel (VDAC2/3), DNA transcription ATP content. These suggest exerts beneficial function mitigate MT2/Akt ST cells.

Testicular dysfunction is often associated with an imbalance in antioxidant/oxidant homeostasis, leading to negative effects of oxidative stress on germ cell proliferation, steroidogenesis, and sperm function. Trace minerals (TM) are involved basic homeostatic enzymatic processes like free radical detoxification, cellular respiration, carbohydrates, lipids, nucleic acids, synthesis metabolism proteins, stabilization membranes DNA.
