Akkermansia muciniphila, a promising candidate for next-generation probiotics, exhibits significant genomic diversity, classified into several distinct clades (AmI to AmIV). Notably, single clade tends predominate within individual hosts, with co-occurrence of different being rare. The mechanisms driving such clade-specific exclusion remain unclear. Here, we show that extracellular vesicles (EVs) derived from AmII inhibit the growth I (AmI), conferring competitive advantage AmII. Moreover, observe immunoglobulin A (IgA) responses, where IgAs, induced by EVs AmII, facilitate niche occupancy and AmI. These findings provide insights dynamics A. muciniphila suggest future personalized microbiome interventions could be optimized considering composition in hosts.

Polyphenols have health-promoting effects, such as improving insulin resistance. Isoxanthohumol (IX), a prenylated flavonoid found in beer hops, has been suggested to reduce obesity and resistance; however, the mechanism remains unknown.High-fat diet-fed mice were administered IX. We analyzed glucose metabolism, gene expression profiles histology of liver, epididymal adipose tissue colon. Lipase activity, fecal lipid plasma metabolomic analysis assessed. Fecal 16s rRNA sequencing was obtained selected bacterial species used for vitro studies. microbiota transplantation monocolonization conducted antibiotic-treated or germ-free (GF) mice. The administration IX lowered weight gain, decreased steatohepatitis improved metabolism. Mechanistically, inhibited pancreatic lipase activity absorption by decreasing fatty acid transporter CD36 small intestine, which confirmed increased excretion feces. markers intestinal barrier function, including thickening mucin layer increasing caludin-1, tight-junction related protein In contrast, effects nullified antibiotics. As revealed using 16S sequencing, microbial community structure changed with significant increase abundance Akkermansia muciniphila IX-treated group. An anaerobic chamber study showed that selectively promoted growth A. while exhibiting antimicrobial against some Bacteroides Clostridium species. To further explore direct effect on we monocolonized either thetaiotaomicron GF mice. jejunum levels multiple classes acids determined analysis. Our demonstrated prevents enhances metabolism inhibiting dietary fat absorption. Constipation is one of the most common gastrointestinal (GI) disorders worldwide. The use probiotics to improve constipation well known. In this study, effect on loperamide-induced by intragastric administration Consti-Biome mixed with SynBalance® SmilinGut (Lactobacillus plantarum PBS067, Lactobacillus rhamnosus LRH020, Bifidobacterium animalis subsp. lactis BL050; Roelmi HPC), L. UALp-05 (Chr. Hansen), acidophilus DDS-1 and Streptococcus thermophilus CKDB027 (Chong Kun Dang Bio) rats was evaluated. To induce constipation, 5 mg/kg loperamide intraperitoneally administered twice a day for 7 days all groups except normal control group. After inducing Dulcolax-S tablets multi-strain were orally once 14 days. 0.5 mL at concentrations 2 × 108 CFU/mL (G1), 109 (G2), 1010 (G3). Compared group (LOP), not only significantly increased number fecal pellets but also improved GI transit rate. mRNA expression levels serotonin- mucin-related genes in colons that treated compared LOP addition, an increase serotonin observed colon. cecum metabolites showed different pattern between probiotics-treated group, short-chain fatty acids probiotic-treated groups. abundances phylum Verrucomicrobia, family Erysipelotrichaceae genus Akkermansia samples Therefore, used experiment thought help alleviate LOP-induced altering acids, serotonin, mucin through improvement intestinal microflora.

Allergic asthma (AA) is a common inflammatory airway disease characterized by increased hyper-responsiveness (AHR), inflammation, and remodeling. Akkermansia muciniphila strictly anaerobic bacterium residing in the gut promising next-generation probiotic to improve metabolic syndrome. A recent study suggested beneficial effect of live A. on allergic inflammation (AAI) mice. However, whether heat-killed form can AAI requires further investigation. Mice sensitized challenged with house dust mites (HDM) develop AA hallmarks including cell infiltration, goblet hyperplasia, subepithelial collagen deposition lungs. These phenomena were reversed oral administration strain EB-AMDK19 (AMDK19-HK) isolated from feces healthy Koreans. Furthermore, AMDK19-HK diminished HDM-induced AHR inhaled methacholine, lung mast accumulation, serum HDM-specific IgE levels. It also led overall suppression IL-4, IL-13, eotaxin production bronchoalveolar lavage fluids, Il4 , Il5 Il13 Ccl17 gene expression tissues. Moreover, suppressed Th2-associated cytokine splenocytes HDM-sensitized mice vitro . Additionally, combination 16S rRNA sequencing short-chain fatty acid (SCFA) analysis cecal samples revealed that modulated relative abundance circulating SCFA-associated genera, positive correlation Lachnospiraceae_ NK4A136_group negative Lachnoclostridium significantly SCFA concentrations. Abstract Osteoarthritis (OA) is a common degenerative joint disease that can cause severe pain, motor dysfunction, and even disability. A growing body of research indicates gut microbiota their associated metabolites are key players in maintaining bone health the progression OA. Short-chain fatty acids (SCFAs) series active widely participate homeostasis. Gold nanoparticles (GNPs) with outstanding anti-bacterial anti-inflammatory properties, have been demonstrated to ameliorate excessive loss during osteoporosis (OP) rheumatoid arthritis (RA). However, protective effects GNPs on OA not clear. Here, we observed significantly alleviated anterior cruciate ligament transection (ACLT)-induced microbiota-dependent manner. 16S rDNA gene sequencing showed changed microbial diversity structure, which manifested as an increase abundance Akkermansia Lactobacillus . Additionally, increased levels SCFAs (such butyric acid), could improved destruction by reducing inflammatory response. Notably, modulated dynamic balance M1/M2 macrophages, serum cytokines such IL-10.
