Osteoporosis is a common metabolic bone disease that results from the imbalance of homeostasis within bone. Intra-bone dependent on precise dynamic balance between resorption by osteoclasts and formation mesenchymal lineage osteoblasts, which comprises series complex highly standardized steps. Programmed cell death (PCD) (e.g., apoptosis, autophagy, ferroptosis, pyroptosis, necroptosis) process involves cascade gene expression events with tight structures. These play certain role in regulating metabolism determining fate cells. Moreover, existing research has suggested natural products derived wide variety dietary components medicinal plants modulate PCDs based different mechanisms, show great potential for prevention treatment osteoporosis, thus revealing emergence more acceptable complementary alternative drugs lower costs, fewer side effects long-term application. Accordingly, this review summarizes types field osteoporosis. perspective targeting PCDs, also discussed roles currently reported osteoporosis involved mechanisms. Based this, provides insights into new molecular mechanisms reference developing anti-osteoporosis future.

In the last decades, ortho-aesthetic-functional rehabilitation had significant advances with advent of implantology. Despite success in implantology surgeries, there is a percentage failures mainly due to loco infections, through bacterial proliferation, presence fungi and biofilm formation, originating peri-implantitis. this sense, several studies have been conducted since then, seeking answers numerous questions that remain unknown. Thus, present work aims discuss interaction between host-oral microbiome development Peri-implantitis was associated diversity species, being Porphiromonas gingivalis, Treponema denticola Tannerella forsythia described higher proportion peri-implantitis samples. parallel role, injury peri-implant tissue causes an inflammatory response mediated by activation innate immune cells such as macrophages, dendritic cells, mast neutrophils. summary, host system may lead imbalance oral microbiota, and, turn, microbiota dysbiosis reported leading cytokines, chemokines, prostaglandins, proteolytic enzymes production. In the field of biomaterials, an endosseous implant is now recognized as osteoimmunomodulatory but not bioinert biomaterial. Scientific advances in bone cell biology and immunology have revealed a close relationship between immune systems resulting science called osteoimmunology. These discoveries allowed for novel interpretation osseointegration representing osteoimmune reaction rather than classic healing response, which activation state macrophages ((M1-M2 polarization) appears to play critical role. The periodontal ligament is located between the bone (alveolar bone) and cementum of tooth, it connected by tough fibers called Sharpey's fibers. Alveolar bone resorption caused by bacteria‐induced periodontitis remains challenging due to sustained inflammation. Periodontal pathogens like Porphyromonas gingivalis launch the primed signal of NOD‐like receptor family pyrin domain‐containing 3 (NLRP3) inflammasome in macrophages; consequent overproduction proinflammatory cytokines and reactive oxygen species (ROS) leads tissue destruction. This provides potential targets for a new therapeutic strategy. Herein, multifunctionalized dual‐crosslinked hydrogel pGM/cPL@NI with NLRP3 inhibitor MCC950 loaded is prepared. Driven strategic functionalization gelatin methacryloyl ε ‐poly‐lysine phenylboronic acid catechol, respectively, containing dynamic photo‐crosslinking networks demonstrates superior mechanical strength stimuli‐responsive behavior, as well overwhelmed performance bacteria killing ROS scavenging. Crucially, restores compromised osteogenesis specifically suppressing cytokine cascade triggered activation promoting anti‐inflammatory polarization macrophages. Collectively, presents robust an effective “cocktail therapy” combining antibacterial, antioxidant, inflammation resolution, regenerative functions. The present study reveals underlying mechanism bacterial‐immune‐regeneration extended approach periodontal engineering.
