Background: Although trauma exposure (TE) is a transdiagnostic risk factor for many psychiatric disorders, not everyone who experiences TE develops disorder. Resilience may explain this heterogeneity; thus, it critical to understand the etiologic underpinnings of resilience. Objective: The present study sought examine genetic resilience using genome-wide association studies (GWAS), complex trait analysis (GCTA), and polygenic score (PRS) analyses.Method: Participants were 6,634 exposed college students attending diverse, public university in Mid Atlantic. GWAS GCTA analyses conducted, summary statistics from large consortia, PRS examined shared between various phenotypes. Results: Results demonstrate that nine single-nucleotide polymorphisms (SNPs) met suggestive significance threshold, heritability estimates non-significant, there overlap AD, as well PTSD.Conclusion: Mixed findings suggest additional research elucidate etiological resilience, ideally with larger samples less biased by variables such heterogeneity (i.e. clinical vs. population based) stratification. Genetic investigations have potential molecular bases stress-related psychopathology, suggesting new avenues prevention intervention efforts.

Background: Based on the gene-environment interaction paradigm, this study explored effect of peer relationships adolescent loneliness and role psychological resilience oxytocin receptor gene (OXTR). Methods: A survey was conducted in a sample 619 adolescents, their oral cells were collected for DNA extraction genotyping. Results: The results showed that (1) both significantly affected loneliness; (2) partially mediated relationship between adolescents; (3) OXTR rs53576 polymorphism moderated first second half indirect pathway mediation model. Specifically, carriers A/A genotype enhanced resilience, while G/G loneliness. Conclusion: These findings helped elucidate developmental mechanisms terms relationships, polymorphisms. The acute stress response is characterized by activation of multiple interconnected systems in the body, resulting release a flood hormones and immune mediators into circulation. In addition to detection these molecules serum, saliva can serve as source markers well be collected non-invasive way. complete profile salivary biomarkers associated with hypothalamic pituitary adrenal/gonadal axes system during has not been fully elucidated. cohort 62 first responders engaged training exercise, we set out determine patterns cytokine, chemokine hormone shifts response. Salivary samples were immediately before (pre-stress), after (post-stress) 1 h test (recovery). Multiplex ELISA panels 42 cytokines 6 steroid thyroid used concentrations three aforementioned time points. Principal components analysis was conducted large data sets collected. our ≥0.3 loading principal analysis, for pre-stress vs. post, post-stress recovery recovery, total three, four factors accounted 56.6, 68.34, 61.70% biomarker variation each phase respectively. ≥0.7 found stages, Of note, MCP1 present all from post-stress, fractalkine pre stress. Additionally, testosterone, estradiol, T4 T3 grouped together consistently same factor phases both analysis. Abstract Resilience or the capacity to “bend but not break” refers ability maintain regain psychobiological equilibrium during after exposure stressful life events. Specifically, resilience has been proposed as a potential resource for staving off pathological states that often emerge repeated stress and are related alterations in circulating cortisol. The aim of this systematic review literature was gather evidence relationship between psychological cortisol levels adult humans. An extensive search carried out following Preferred Reporting Items Systematic Reviews Meta‐Analyses (PRISMA) method PubMed Web Science databases. In total, 1256 articles were identified and, these, 35 peer‐reviewed included review. We categorized findings according (1) short long‐term secretion period covered by matrices selected studies also (2) differentiated diurnal, phasic (acute), tonic (basal) components HPA output which they refer their relationships with resilience. Reported distinct parameters varied widely across studies, finding positive, negative, null associations two variables. Notably, several found no used single morning saliva plasma sample assessment axis activity. Despite limitations such great variability instruments methods measure both cortisol, together high heterogeneity small sizes, points modifiable key factor modulate physiological response stress. Therefore, further exploration interaction variables is necessary eventual development future interventions aimed at promoting an essential component health prevention.

Introduction: Posttraumatic stress disorder (PTSD) is a complex multifactorial influenced by the interaction of genetic and environmental factors. Analyses epigenomic transcriptomic modifications may help to dissect biological factors underlying gene-environment interplay in PTSD. To date, most human PTSD epigenetics studies have used peripheral tissue, these findings poorly understood relationships brain alterations. Studies examining tissue characterize brain-specific profiles In this review, we compiled integrated molecular from humans animals. Methods: A systematic literature search according PRISMA criteria was performed identify PTSD, focusing on postmortem samples or animal-stress paradigms. Results: Gene- pathway-level convergence analyses revealed PTSD-dysregulated genes pathways across regions species. total 243 converged species, with 17 them significantly enriched for Chemical synaptic transmission signaling G-protein-coupled receptors were consistently omics Discussion: Our point out dysregulated highly replicated animal models suggest potential role corticotropin-releasing hormone/orexin pathway PTSD’s pathophysiology. Further, highlight current knowledge gaps limitations recommend future directions address them.

In this review, we present selection some the most relevant data on translational biomarkers in field mental Several critical aspects concerning preclinical clinical model development are addressed. distance between disease widened with time across all fields medicine, psychiatry presenting additional hurdles represented by inherent heterogeneity studied phenotypes. Capitalizing technological advances developing consolidating sound theories stress-resilience represents avenue, possibly endowed greater ecological validity compared to sole socio-psychological assessment. Instrumental advancing will be an increased level integration researchers' efforts biomarkers, aiming elucidate better interindividual impact exposure individuals' health behavior.

Stress-induced conditions are associated with impaired cerebral blood flow (CBF) and increased risk of dementia stroke. However, these do not develop in resilient humans animals. Here the effects predator stress (PS, cat urine scent, ten days) on CBF mechanisms regulation were compared PS-susceptible (PSs) PS-resilient (PSr) rats. Fourteen days post-stress, rats segregated into PSs PSr groups based a behavior-related anxiety index (AI). its endothelium-dependent changes measured parietal cortex by laser Doppler flowmetry. The major findings are: (1) PS susceptibility was reduced basal endothelial dysfunction. In rats, higher, dysfunction attenuated. (2) inversely correlated AI PS-exposed (3) Endothelial decrease eNOS mRNA to control (4) Brain dopamine (5) Plasma corticosterone (6) A hypercoagulation state present but Thus, potential resilience that protective for identified.
