Abstract Heavy metals are naturally occurring components of the Earth’s crust and persistent environmental pollutants. Human exposure to heavy occurs via various pathways, including inhalation air/dust particles, ingesting contaminated water or soil, through food chain. Their bioaccumulation may lead diverse toxic effects affecting different body tissues organ systems. The toxicity depends on properties given metal, dose, route, duration (acute chronic), extent bioaccumulation. detrimental impacts human health largely linked their capacity interfere with antioxidant defense mechanisms, primarily interaction intracellular glutathione (GSH) sulfhydryl groups (R-SH) enzymes such as superoxide dismutase (SOD), catalase, peroxidase (GPx), reductase (GR), other enzyme Although arsenic (As) is believed bind directly critical thiols, alternative hydrogen peroxide production processes have also been postulated. known signaling pathways affect a variety cellular processes, cell growth, proliferation, survival, metabolism, apoptosis. For example, cadmium can BLC-2 family proteins involved in mitochondrial death overexpression antiapoptotic Bcl-2 suppression proapoptotic (BAX, BAK) thus increasing resistance cells undergo malignant transformation. Nuclear factor erythroid 2-related 2 (Nrf2) an important regulator enzymes, level oxidative stress, oxidants has shown act double-edged sword response arsenic-induced stress. Another mechanism significant threats metal (e.g., Pb) involves substitution essential calcium (Ca), copper (Cu), iron (Fe)) structurally similar (Cd) (Pb)) metal-binding sites proteins. Displaced redox (copper, iron, manganese) from natural catalyze decomposition Fenton reaction generate damaging ROS hydroxyl radicals, causing damage lipids, proteins, DNA. Conversely, some metals, cadmium, suppress synthesis nitric oxide radical (NO · ), manifested by altered vasorelaxation and, consequently, blood pressure regulation. Abstract Mercury (Hg) is a widely distributed and bioavailable metal of public health concern, with many known human toxicities, but data regarding mercury's influence on thyroid cancer (TC) scarce. to impact several molecular pathways implicated in carcinogenesis, its proclivity for bioaccumulation the suggests potential modulatory effect. We conducted literature/systematic review studies between 1995–2022 intending define better establish relationships these two entities, congregate evidence role identify populations interest further study. Insufficient precludes definitive conclusions dietary mercury as TC risk factor; however, common mechanisms affected by are crucial development, including biochemical, endocrine, reactive oxygen species effects. Quantitative analysis revealed associations exposure. In three studies, average urine levels were higher patients, mean difference 1.86 µg/g creatinine (95% CI = 0.32–3.41). investigating exposure elevated levels, exposed group exhibited developing TC, relative 1.90 1.76–2.06). tissue (ppm) averaging 0.14 (0.06–0.22) cancerous cases (N 178) 0.08 (0.04–0.11) normal thyroids 257). Our findings suggest an association risk, implying possible predisposing factor. Further research necessary reveal clinical relevance environmental exposures pathogenesis.

Oxidative stress occurs when there is an imbalance between the production of reactive oxygen species (ROS) and body’s antioxidant defenses. It poses a significant threat to physiological function reproductive cells. Factors such as xenobiotics heat can worsen this stress, leading cellular damage apoptosis, ultimately decreasing efficiency. The nuclear factor erythroid 2–related 2 (Nrf2) signaling pathway plays crucial role in defending against oxidative protecting cells via enhancing responses. Dysregulation Nrf2 has been associated with infertility suboptimal performance mammals. Cadmium has been well recognized as a critical toxic agent in acute and chronic poisoning cases occupational nonoccupational settings environmental exposure situations. is released into the environment after natural anthropogenic activities, particularly contaminated industrial areas, causing food pollution. In body, cadmium no biological activity, but it accumulates primarily liver kidney, which are considered main targets of its toxicity, through oxidative stress inflammation. However, last few years, this metal linked to metabolic diseases. The pancreas-liver-adipose axis largely affected by accumulation. Therefore, review aims collect bibliographic information that establishes basis for understanding molecular cellular mechanisms with carbohydrate, lipids, endocrine impairments contribute developing insulin resistance, syndrome, prediabetes, diabetes.

Introduction: Cadmium (Cd) is a harmful heavy metal that results in many toxic issues. Urtica pilulifera showed potential pharmaceutical applications. This study investigated the possible ameliorative mechanism of leaves extract (UPLE) against hepatotoxicity induced by cadmium chloride (CdCl 2 ) mice. Methods: In vitro phytochemical screening and metal-chelating activity UPLE were ascertained. Four groups forty male mice used ( n = 10) as follows; Group 1 (G1) was negative control. G2 injected i.p., with (100 mg/kg b. wt) daily. G3 Cd (5 G4 G2. On day 11, body weight changes evaluated, blood, serum samples collected for hematological biochemical assessments. Liver tissues biochemical, molecular, histopathological investigations. Results: The contains promising secondary metabolites considerably lessen effects on liver. Furthermore, inhibited oxidative stress inflammation; restored antioxidant molecules; promoted nuclear-related factor-2 (Nrf-2) expression. Also, improved alterations Cd. Discussion: explored beneficial role treatment Cd-induced liver injury through enhancing Nrf-2 signaling enzyme gene expression Therefore, could have valuable implications environmental exposure.
